STAT3\PCNA在胎盘组织中的表达及与胎盘老化的相关性研究

【摘要】 目的分析凋亡相关蛋白STAT3及增殖蛋白PCNA在胎盘发育过程中的表达及功能,探讨二者与胎盘老化的相关性。方法采用免疫组织化学方法分别检测妊娠不同阶段胎盘组织及老化胎盘组织中STAT3和PCNA蛋白的表达。结果①STAT3、PCNA在细胞滋养细胞中表达,在合体滋养细胞中不表达;②STAT3的表达强度在早期和晚期妊娠组间差异有极显著性。③PCNA表达强度在妊娠早、晚期组间差异有极显著性;中、晚期组间差异有显著性;④STAT3及PCNA表达在正常妊娠各组间有显著正相关性;在老化组无相关性。结论①PCNA在妊娠早、中期高度表达与滋养细胞的高度增殖状态及旺盛的功能相关;②STAT3在妊娠早、中期表达量较高,可能通过抑制滋养细胞的凋亡而维持滋养细胞功能,进而维持妊娠;③胎盘老化可能与STAT3的表达失活有关;④正常妊娠各组STAT3与PCNA表达呈显著正相关,说明STAT3可能通过抑制滋养细胞的凋亡而影响细胞的增殖,从而使得细胞滋养细胞保持增殖与凋亡的平衡状态,保证胎盘功能的正常运行。

【Abstract】 【Objective】To study the expression and function of STAT3 and PCNA in normal and aging placenta and discuss the relationship between STAT3\PCNA and placenta aging.【Method】Immunohistochemistry method was used to examine the expression of PCNA and STAT3 in 42 placentas of all pregnancy periods.【Result】PCNA and STAT3 were not found in syncytiotrophoblasts(s-cells).Significant difference was found between mid and late pregnancy.the most significant difference was found between early and late pregnancy.The positive rate of STAT3 was different between early and late pregnancy.The expressive intensity of STAT3 was significantly different be-tween early and late pregnancy.There were positive relations between PCNA and STAT3 expression in normal preg-nancy.No relations were observed in STAT3 and PCNA expression in late pregnancy and aging group.【Conclu-sion】 The high expression of PCNA illustrates that PCNA relates to trophocytes’ high proliferation and endocrine function in early and mid pregnancy.During the development of placenta in early and mid pregnancy,STAT3 sup-presses trophocyte apoptosis to sustain pregnancy.Placenta aging relates to the deactivation of STAT3.The positive relationship between STAT3 and PCNA in normal pregnancy shows that STAT3 may influence trophocyte proliferat-ing and sustain placenta’ function by suppressing trophocyte apoptosis.
