血小板源生长因子B及其受体β与支气管哮喘血管再生与重塑
Platelet derived growth factor B and its receptor β in airway vascular angiogenesis and remodeling in a murine model of bronchial asthma
【摘要】 目的进一步明确支气管哮喘时血管相关因子血小板源生长因子B(PDGFB)和血小板源生长因子受体β(PDGFRβ)在支气管哮喘小鼠模型气管及肺组织中的变化和作用。方法在建立小鼠支气管哮喘模型的基础上,应用免疫荧光染色和HE染色计数血管平滑肌细胞数量变化,应用逆转录多聚酶链反应(RT-PCR)方法检测气管和肺组织PDGFB及其受体(PDGFB/PDGFRβ)mRNA表达情况,应用Western blot方法检测PDGFB/PDGFRβ蛋白水平。结果①血管平滑肌细胞数量在支气管哮喘急性期减少,在慢性期又恢复正常;②应用RT-PCR技术及琼脂糖凝胶电泳,PDGFB mRNA及PDGFRβ mRNA在正常气管未检测到,而在慢性延长期哮喘小鼠气管组织PDGFB mRNA显示出清晰的条带。周围肺组织在正常小鼠也表达PDGFB mRNA,哮喘小鼠肺组织表达增强,在慢性延长期表达明显增加。应用Western blot蛋白检测技术,哮喘小鼠气管及肺组织PDGFB及PDGFRβ蛋白表达情况与基因检测结果相一致。结论PDGFB和PDGFRβ基因与蛋白表达水平在支气管哮喘气管及肺组织均增加,特别在慢性延长期,PDGFB和PDGFRβ表达改变是血管平滑肌细胞数量变化的重要因素。PDGFB和PDGFRβ系统在支气管哮喘气道血管再生和重塑过程中可能起着重要的作用。
【Abstract】 Objective To elucidate the role of platelet derived growth factor B(PDGFB) and platelet derived growth factor receptor β(PDGFRβ) in vascular angiogenesis and remodeling in the mouse model of asthma.Methods After establish ment of the mouse model of bronchial asthma,immunofluorescence and HE staining were used to count the number of vascular smooth muscle cells(vSMC),then reverse transcription-polymerase chain reaction(RT-PCR) and western blot methods were used to detect mRNA and protein level of PDGFB and PDGFRβ in trachea and lung tissues.Results ①Smooth muscle cells decreased in acute phase,however,they returned to nearly normal in chronic stage;②PDGFB mRNA and PDGFRβ mRNA were not detected in normal tracheal tissue.But they were increased in trachea in chronic asthma.They showed enhanced expressions in asthma lung tissues compared with those in normal tissues,especially in chronic phase.The results of protein levels using western blot method were consistent with mRNA expression.Conclusions PDGFB and PDGFRβ mRNA and protein showed enhanced expressions in asthma tracheal and lung tissues compared with those in normal tissues,especially in chronic phase.This is a main factor of vSMC changes.PDGFB and PDGFRβ may play an important role in vascular angiogenesis and remodeling in asthma murine model.
