血浆同型半胱氨酸水平对结直肠癌和肝细胞癌细胞增殖的影响

【摘要】 目的:探讨同型半胱氨酸(Hcy)对结直肠癌和肝癌增殖的影响。方法:收集结直肠癌、肝癌和健康人血浆204份测定Hcy。再检测Hcy对HepG2和HT29增殖的影响。结果:癌症组Hcy水平与健康组比较有统计学差异(P<0.05)。HepG2在4和2 mmol/L Hcy处理组与未处理组比较OD值增高(P<0.05);0.5和0.25 mmol/L处理组与未处理组比较OD值减低(P<0.05)。HT29在4 mmol/L Hcy处理组与未处理组比较OD值增高(P<0.05)。HepG2经0.5和0.25 mmol/L Hcy处理,DNA凝胶电泳均未形成梯状图形。HepG2和HT29经4和1 mmol/L Hcy处理,S期细胞构成比与未处理组比较均无统计学差异(P>0.05)。HepG2经0.25 mmol/L Hcy处理未检出亚二倍体峰。结论:Hcy达一定浓度时有促进肿瘤细胞增殖的潜能。

【Abstract】 Objective:To investigate the influence of homocysteine(Hcy) in proliferation of colorectal cancer and liver cancer.Methods:Hcy status of patients suffering with colorectal cancer or liver cancer was separately compared with that of normal control group.Then we used MTT method,DNA gel electrophoresis and flow cytometry to examin the effect of various doses of Hcy in tumor cell lines HepG2 and HT29.Results:Mean Hcy level in colorectal cancer and liver cancer were significantly higher than that in healthy control group(P<0.05).In HepG2,the OD values of 4 mmol/L and 2 mmol/L Hcy groups were significantly higher than that of negative control group(P<0.05);but the OD values of 0.5 mmol/L and 0.25 mmol/L Hcy groups were significantly lower than that of negative control group(P<0.05).In HT29,the OD values of 4 mmol/L Hcy group were also significantly higher than that of negative control group(P<0.05).By DNA gel electrophoresis,we did not find ladder straps in 0.5 mmol/L and 0.25 mmol/L Hcy groups of HepG2.By flow cytometry,the results showed that in HepG2 and HT29 the percentage of cells in S phase had no difference between various doses groups(4 mmol/L,1 mmol/L Hcy) and negative control group(P>0.05).And in HepG2,we did not find sub-population in 0.25 mmol/L Hcy group.Conclusion:Hcy have the potential of promoting tumor cells growth at certain concentrations.
