高效液相色谱法快速测定黄体酮血药浓度

【摘要】 目的建立高效液相色谱法快速测定人血浆中黄体酮血药浓度。方法色谱柱为Hypersil ODS2(5μm,4.6mm×250mm);流动相为乙腈:5mmol/L三羟甲基氨基甲烷(pH=8.0)=63:37;柱温:室温;流速1.0ml/min;检测波长:240nm。结果本测定方法的线性范围:10~1600ng/ml,r=0.9998。平均回收率为(96.1±3.1)%,日内RSD≤5.7%,日间RSD≤6.4%。黄体酮的最低检测浓度为6ng/ml。结论本方法简单、快速、灵敏、重现性好,适用于黄体酮临床血药浓度监测及人体药代动力学研究。

【Abstract】 Objective To establish a rapid HPLC method for determination of progesterone in plasma. Methods A Hypersil ODS2(5 μm,4.6 mm×250 mm) was adopted. Mobile phase was a mixture of CH3CN and 5 mmol/L Tri(hydroxymethyl)aminomethane (pH=8.0)(63:37).The flow rate was 1.0 ml/min. The detection wavelength of progesterone was at 240 nm . Results The assay procedure was shown to produce linear calibration curves over the range of 10 ng/ml to 1600 ng/ml of progesterone in plasma (r=0.9998). Within the range, the recovery rate was (96.1±3.1)% , and the RSD within a day and between days was less than 5.7% and 6.4% ,respectively.The detection limit of progesterone was 6 ng/ml.Conclusions This method is found to be reproducible, convenient and sensitive for plasma concentration monitoring and clinical pharmacokinetics study of progesterone
