脂质体转染Akt基因对心肌细胞缺血-再灌注的影响
Experimental Study of Cationic Liposome Mediated Transgenic Human Akt Gene into Cardiomyocytes and Its Protective Effect on Ischemia/Reperfusion Injury in Vitro
【摘要】 目的研究转染脂质体介导的Akt基因对心肌细胞缺血-再灌注的影响。方法采用新生SD大鼠进行心肌细胞培养,建立模拟缺血/再灌注模型,将脂质体介导的人Akt基因转染心肌细胞。实验分5组:对照组(control组)、缺血/再灌注组(I-R组)、转染Akt基因组(Akt组)、转染阳离子脂质体空载体组(Vehicle control组)和Akt抑制剂组(Akt blockade组)。各组进行缺血-再灌注后以MTS比色法检测细胞活性,测定乳酸脱氢酶(LDH)活性、丙二醛(MDA)含量。结果Akt组较I/R组、Vehicle control组和Akt blockade组LDH、MDA明显降低,细胞活性高(P<0.05)。结论脂质体介导的Akt基因转染心肌细胞可保护心肌、减轻心肌细胞缺血-再灌注损伤。
【Abstract】 Objective To explore the effect and mechanism of cationic liposome mediated transgenic human Akt gene against ischemia/reperfusion (I/R) in cardiomyocytes in vitro. Methods Cardiomyocytes from new born Sprague-Dawley rats were isolated and cultured in vitro and were randomly divided into five groups:control group ,ischemia/ reperfusion (I/R) group ,Akt group ,vehicle control group and Akt blockade group. The cell activity and the levels of lactate dehydrogenase (LDH) in the medium ,malondialdehyde (MDA) in myocardiocytes were tested. Results In comparison with I/R group,vehicle control group and Akt blockade group ,the levels of LDH and MDA in Akt group were significantly decreased (P<0.05),together with higher cell activity (P<0.05).Conclusion Cationic liposome mediated transfection of Akt gene can protect cultured cardiomyocytes against ischemia/reperfusion injury.
