两种采样法在海、水产品污染霍乱弧菌检测结果中的比较分析
Comparison of difference of detecting Bacillus comma in marine and fishery products by two sampling methods
【摘要】 目的:比较涂抹法与整体或部分采样法在海、水产品检测霍乱弧菌结果中的差异,推广简便高效的采样方法,提高霍乱监测的灵敏度。方法:同种样品同时使用两种采样方法,采集海、水产品进行增菌、分离、鉴定;阳性菌再做霍乱ctxA基因检测。结果:512份样品经涂抹采样法检出霍乱弧菌O1群26株,检出率为5.08%(26/512);整体或部分采样法检出霍乱弧菌O1群16株,检出率为3.13%(16/512)。ctxA基因检测均为阴性。经卡方检验,两种采样方法在海、水产品检测霍乱弧菌的总体、分类检出率均没有显著性差异(P>0.05);但鱼类的检出率两者有显著性差异(2χ=3.88,P<0.05),涂抹采样法检出率较高,而且检出阳性样品种类比常规法多。结论:两种采样法在现场考核中都切实可行,行之有效,但涂抹采样法更具优势,效果更显著;建议在上述海、水产品的霍乱弧菌监测和检测中应首选涂抹采样法,尤其是鱼类。
【Abstract】 Objective:To compare the difference of detecting Bacillus comma in the marine and fishery products by the painting and the whole or part sample drawing.To select and generalize which one is convenient and high-performance method to elevate sensitivity of detecting Bacillus comma.Methods:Apply the two methods,gather the marine and fishery products and then to cultured,isolated and identified Bacillus comma.Then choise the positive samples and detected ctxA gene of Bacillus comma.Results:For the 512 samples,we detected 26 strains of Bacillus comma O1 by painting method,and the detection rate is 5.08%(26/512);16 strains were detected by the whole or part sample drawing method,and the detection rate is 3.13%(16/512).The detection of ctxA gene is negative by two methods.It is not different to detect Bacillus comma′s total and categorial detection rate of by the two sample drawing in the marine and fishery products by the method of the χ2 test(P>0.05);But in ichthy-products,we could find the besmear sample drawing higher than the whole or part sample drawing(χ2=3.88,P<0.05).Conclusion:The two sampling methods are all practicable and effective.And the painting sampling method has more advantages and more effective.So we suggest that the first choice be painting sampling method,particularly in fish.
