心肌细胞钙瞬变共聚焦显微图像的处理分析

【摘要】 目的分析心肌细胞钙瞬变共聚焦图像的特点和处理过程。方法心肌细胞钙瞬变由电刺激激发,钙信号指示剂为Fluo-4,其图像由激光共聚焦显微镜记录;之后选用适当的软件和自编的一些程序(IDL语言),对原始图像进行格式转换、标准化后,提取其特征曲线和重要参数,最后与重构的钙瞬变图像一起呈现。结果钙瞬变图像经上述过程处理后,其所包含的在钙瞬变过程中胞内胞信号随时间的变化过程和细胞长度随时间缩短或恢复的过程等重要信息得到有效提取。此两类重要信息与重构后的钙瞬变图像一起呈现使结果完整、直观、明确。结论采用本文提出的方法处理分析心肌细胞钙瞬变图像可达到提取重要信息、明确实验结果的目的,为进一步的统计分析奠定了基础。

【Abstract】 Objective To analyze the features of calcium transient images from cardiomyocytes and the image processing mothods.Methods Cardiomyocytes calcium transients were triggered by electric stimulus.Using Fluo-4 as the calcium indicator,the images were collected by a laser scanning confocal microscope.Using some softwares and self-coded programs(with IDL),the featured curves and some key parameters were extracted after image format transfer and normalization.All the results were displayed with reconstructed images of the calcium transients.Results After image processing,key information about the intracellular calcium signal and cellular length based on time during calcium transients were obtained.These two types of important information along with the reconstructed image made a complete,distinct and clear results.Conclusions The image processing and analysis introduced in the present study make it possible to obtain key information from cardiomyocytes calcium transients and to reach a clear result.It is very helpful for further data statistics.
