血管腔内β-照射对预防冠状动脉再狭窄的动物实验研究
Intracoronary β-Radiation in Preventing Restenosis of Balloon-Injured Coronary Artery in Pigs
【摘要】 目的探讨冠状动脉球囊扩张后腔内β-照射预防血管再狭窄的意义。方法12头猪的冠状动脉左前降支(LAD)和左回旋支(LCX)用球囊导管予以扩张,其中6头LAD和LCX用后装放疗装置分别给予32磷15-20 Gy的腔内照射,其余6头作为对照组。28天后切取损伤部位的冠状动脉,石腊包埋、切片。用计算机辅助组织形态学测量仪进行测量。结果照射组血管腔面积为LAD 3.98±0.79 mm2和LCX 4.08±0.59 mm2,对照组则分别为LAD 2.54±0.61mm2和LCX 3.01±0.55 mm2(P<0.05)。新生内膜面积照射组为LAD 1.21±0.27 mm2和LXC 1.19±0.23 mm2,对照组为LAD 1.67±0.18 mm2和LCX 1.73±0.14 mm2(P<0.01)。结论猪的冠状动脉球囊扩张后,经腔内β-照射可抑制新生内膜的形成,增加血管腔面积从而预防血管的再狭窄。
【Abstract】 Objective The purpose of this study is to investigate the effects of intracoronary β-radiation in preventing restenosis.Methods 12 pigs underwent balloon-angioplasty to cause an overstretching injury on LAD and LCX.Six pigs received 32P radiation with a does of 15 or 20 Gy and remainder were not irradiated(controls).32P was delivered into a catheter via an afterloading device on balloon-injured coronary arteries in pigs.Sections of paraffin embedded injured arteries,perfusion fixed at 4 weeks,were measured by computer-assisted histomorphometry.Luminal area was 3.98±0.79 mm2(20 Gy) and 4.08±0.59 mm2(15 Gy) compared to 2.54±0.61 mm2 and 3.01±0.55 mm2 in the controls(P<0.05).Neointimal area was 1.21±0.27 mm2(20 Gy) and 1.19±0.23 mm2(15 Gy) compared to 1.67±0.18 mm2 and 1.73±0.14 mm2 in the controls(P<0.01).Results Conclusion Intracoronary β-radiation significantly increased lumen size and inhibited neointima formation in balloon-injured coronary arteries 28 days after the interventional procedure.
