重组EBV-TK及DNase检测鼻咽癌血清相应抗体的比较研究

【摘要】 【目的】比较鼻咽癌(NPC)患者血清中EB病毒(EBV)胸苷激酶(TK)和DNA酶(DNase)抗体水平。【方法】用鼻咽癌血清中和Raji细胞产生的EBV DNase,抗酶率(AER)≥30%为EBV DNase抗体(EDAb)阳性。以大肠杆菌表达的重组EBV TK为抗原,用Western blot方法检测鼻咽癌血清中的EBV TK抗体(ETAb),显现棕褐色条带者为ETAb阳性。【结果】55例鼻咽癌患者血清,EDAb阳性率94.5%,ETAb阳性率89.1%,EDAb~+/ETAb~+、EDAb~-/ETAb~+、EDAb~+/ETAb~-、EDAb~-/ETAb~-的检出率分别为87.3%、1.8%、7.3%和3.6%,同时检测此两项指标可使EBV相关抗原抗体的阳性率提高至96.4%。【结论】大多数鼻咽癌血清均同时具有EDAb和ETAb,检测这两个酶的抗体是鼻咽癌临床辅助诊断的二个重要血清学指标,联合应用意义更大。

【Abstract】 [Objective]To compare the titer of antibody against EBV TK with titer of antibody against EBV DNase in sera of patients with nasopharyngeal carcinoma(NPC).[Methods]EBV DNase produced by Raji cells was neutralized by the sera of NPC patients and the anti-enzyme rate(AER)≥30%was defined as a cut-off point between positive and negative of EBV DNase antibody(EDAb).The recombinant EBV TK expressed in E.coli was used as an antigen to detect the antibody against EBV TK (ETAb) by Western blot.[Results]In 55 serum samples from NPC patients positive rates of EDAb~+,ETAb~+,EDAb~+/ETAb~+, EDAbVETAb~+,EDAb~+/ETAb~-,EDAb~-/ETAb~- were 94.5%,89.1%,87.3%,1.8%,7.3%,and 3.6%,respectively.The positive rate of antibody against EBV related proteins increased to 96.4%when ETAb and ETAb were detected simultaneously. [Conclusion]EDAb and ETAb were detectable in the sera of most NPC patients and could be used as two serological indices for NPC diagnosis.It would be more valuable to measure both EDAb and ETAb instead of any one of them.
