应用搅拌式生物反应器大量扩增人胎盘间充质干细胞的实验研究
Large Scale Expansion of Human Placenta-derived Mesenchymal Cells in Stirring Bioreactor
【摘要】 探讨应用搅拌式生物反应器培养技术体外扩增人胎盘间充质干细胞(hPDMCs)的方法。以hPDMCs为种子细胞,利用微载体悬浮法在搅拌式生物反应器内进行体外培养,同时设置静态培养的培养瓶为对照组,观察细胞的扩增速率、细胞代谢(乳酸、葡萄糖、培养液的pH值)的变化,检测生物反应器培养前、后干细胞的表面标记物。hPDMCs在搅拌式生物反应器内每代可以扩增(10.55±1.62)倍,明显高于对照组(6.10±0.11)倍的扩增值(P<0.05),且细胞生长代谢指标优于对照组;流式细胞仪检测应用搅拌式生物反应器,培养后细胞表面标记物的表达率无明显改变(P>0.05)。搅拌式生物反应器能够提供良好的细胞生长环境,建立用于hPDMCs的大量扩增的体外三维培养体系。
【Abstract】 This study is aimed to expand human placenta-derived mesenchymal stem cells(hPDMCs) in a stirring bioreactor.hPDMCs were cultured on cyterdex-3 microcarrier in a stirring bioreactor,static culture flask for the control groups. The velocity of expansion and metabolism indexes(lactic acid,glucose,pH in medium) were detedcted.The surface marker of hPDMCs was detected by FACS.hPDMCs were expanded(10.55±1.62) times cultivated in the stirring bioreactor per passage,markedly surpassed that of the control groups(6.10±0.11) times(P<0.05),and there was no significant change of the surface markers of hPDMCs cultured in the stirring bioreactor(P>0.05).The stirring bioreactor could provide a better environment for hPDMCs expansion,showing great potential to be a 3D culture system for hPDMCs expansion in vitro.
