红外光谱法研究极低频电磁场暴露对大鼠肺组织二级结构的影响

【摘要】 为了探讨极低频电磁场长期暴露对大鼠肺组织结构改变机理,采用傅里叶变换红外(FTIR)光谱法研究暴露/假暴露于电场强度E=3.5~4 kV/m,磁场强度B=8~10μT的极低频电磁场400 d后大鼠肺组织红外谱和二维相关红外光谱,根据大鼠肺组织傅立叶变换红外光谱酰胺I带的拟合分析,获得组织中蛋白质二级结构数目及其组成。结果表明:极低频电磁场暴露引起酰胺I带峰产生位移;暴露组蛋白质二级结构稳定性下降,说明极低频电磁场长期暴露引起了肺组织中蛋白质变性;二维相关红外谱的变化规律同红外谱结果一致,进一步证明暴露组大鼠肺组织发生的主要变化是蛋白质变性以及二级结构组成发生改变。本实验为在分子光谱水平上解释极低频电磁场生物效应提供了一种研究手段。

【Abstract】 Recently,great attention has been paid to the effects of exposure to extremely low frequency electromagnetic fields(ELF EMFs) on human being,since there are many devices and environment that generate electromagnetic fields.In this study,the effects of ELF EMFs on male rat lung tissues were investigated via Fourier transform infrared(IR) spectrums and two-dimensional infrared(2D-IR) correlation spectroscopy with experimental animals under the exposure to E=3.5~4 kV/m,B=8~10 μT for 400 days.Considerable different compositions of the secondary structure of protein,determined by curve-fitting analysis of the amide I bands in the FTIR spectra,were found in lung tissues between exposed group and sham-exposed group,suggesting that amideⅠabsorption peak of exposed group shifted slightly compared with sham-exposed group.Moreover,ELF EMFs seemed to affect the protein secondary structure,especially the ordered secondary structure of the exposed group decreased markedly by analyzing the components of secondary structure of amide I bonds,which indicated that the denaturation occurred in protein compound under longtime exposure to ELF EMFs.In addition,2D IR correlation spectroscopy showed the same results.The IR spectroscopy method can track dynamically the process of exposure to ELF EMFs and reveal mechanism of bio-effects of ELF EMFs.
