响应面法优化超声提取紫玉米苞叶花青素工艺条件的研究

【摘要】 花青素是一类广泛存在于植物界,色泽鲜艳、无毒、呈现红色至蓝色的色素,紫玉米茎、叶、果实中含有大量的花青素,来源广泛,价格低廉,是提取花青素的良好来源,紫玉米苞叶中花青素含量尤其丰富,且大部分被用做饲料,浪费严重。传统的提取方法提取费时,提取成本高。超声辅助提取具有方便、快速、简单、安全等优点。故本试验利用响应面法对超声提取紫玉米苞叶花青素的工艺进行了优化,分别对超声时间、料液比、乙醇浓度进行响应面分析,得出提取紫玉米苞叶花青素的最佳工艺条件为:超声时间24.98min,料液比1∶30g/mL,乙醇浓度79.25%,花青素得率为3507.7mg/100g。实际验证花青素得率为3498.6mg/100g,与预测值相对误差为0.26%,符合实际生产需求,比单因素的最佳组合提取率提高了8.67%。

【Abstract】 Anthocyanins are a class of widely exist in the vegetable kingdom,the brightly-colored,non-toxic,showed red to blue pigment.Purple corn stems,leaves,fruits contain many Procyanidins,a broad range of sources and low prices,are a good source of extraction of anthocyanins,purple corn bract especially rich in anthocyanin content,and is used to do the majority of feed,waste.Traditional extraction methods extract time-consuming,high cost of extraction.Ultrasound-assisted extraction with a convenient,fast,easy,safe,etc.The process of ultrasound extraction anthocyanins from purple corn leaf was optimized by the method to response surface.Three process parameters including ultrasound time feed ratio,alcohol concentration were analized,respectively.As a result,the best process of extraction anthocyanins from purple corn leaf was as follows:extrative time 24.98min,extractive temperature 60℃,feed ratio 1:30g/mL,alcohol concentration 79.25%.Doing like that,the ratio of anthocyanins preservation was 3498.6mg/100g,3507.7mg/100g predictive value and relative error of 0.26%,in line with actual production needs increasing 8.67% than that of the best single facters combination.
