激活素结合蛋白在ConA诱导小鼠肝损伤时的表达及作用

【摘要】 目的:探讨激活素结合蛋白(Follistatin,FS)在刀豆蛋白A(ConcanavalinA,Con A)诱导肝损伤中的表达及作用。方法:尾静脉注射ConA诱导小鼠急性肝损伤,实时定量PCR检测FSmRNA表达,ELISA法检测FS及激活素A(Activin A)蛋白水平。结果:注射ConA后肝脏组织中FSmRNA表达水平在第三天明显下降,FS蛋白水平在第三天也呈下降趋势;而Activin A水平明显增高。注射FS中和内源性激活素A作用后,肝脏病理损伤程度明显减轻,血清转氨酶水平也显著低于ConA单纯诱导组。结论:在ConA诱导的急性肝损伤过程中激活素A-FS系统失平衡,应用FS阻断内源激活素A可以减轻ConA诱导的肝脏损伤,因此FS有可能成为治疗肝损伤性疾病的有效药物。

【Abstract】 Objective:To investigate the expression and roles of activin-binding protein(follistatin,FS)in ConA-induced mouse liver injury.Methods:Acute liver injury of mice was induced by injection with ConA intravenously via tail vein.FS mRNA expression was examined by real-time quantitative PCR.FS and activin A protein levels were detected by enzyme-linked immunosorbent assay(ELISA).Results:FS mRNA levels obviously decreased 3 days after ConA administration,and FS protein levels slightly declined,whereas activin A levels significantly increased.The blockade of activin A action by FS administration attenuated the hepatic injury and reduced serum transaminase levels significantly.Conclusion:The imbalance between activin A and FS may contribute to ConA-induced hepatic injury.Thus,blockade of activin A actions by FS may provide effective way for treatment of liver diseases.
