尿酸对肾小管上皮细胞人尿酸盐转运子基因表达的影响
Effect of uric acid on the expression of human urate transporter gene in HK-2 cells
【摘要】 目的:观察不同浓度尿酸是否调节肾小管上皮细胞(HK-2细胞)人尿酸盐转运子(hUAT)mRNA的表达。方法:根据培养液中所含尿酸浓度的不同,将HK-2细胞分为:①仅使用DMEM(对照组);②DMEM+100μmol/L尿酸(U1组);③DMEM+200μmol/L尿酸(U2组);④DMEM+400μmol/L尿酸(U3组)。每组均培养6瓶细胞。上述细胞在不同培养液中分别培养48小时。采用实时荧光定量PCR检测HK-2细胞中hUAT mRNA的相对表达量(2-ΔΔCt法)。结果:所有标本均能检测到hUAT mRNA的表达,且各组hUAT mRNA表达水平均明显低于对照组,U1组hUAT mRNA表达量为对照组的108%,U2组为对照组的111%,U3组为对照组的116%。除U1组外各组与对照组相比,差异均有显著性(P<0.05),但各组间差异均无统计学意义(P=0.49,0.54,0.15)。结论:尿酸可上调hUAT mRNA表达。
【Abstract】 Objective:To observe the effect of uric acid in different concentrations on the expression of human urate transporter(hUAT) gene in HK-2 cells.Methods:HK-2 cells were divided into control(cultured with DMEM),U1(DMEM+100 μmol/L uric acid),U2(DMEM+ 200 μmol/L uric acid),and U3(DMEM+400 μmol/L uric acid) groups.In each group,6 bottles of the cells were cultured in vitro for 48 hours.Then the relative quantity of hUAT mRNA in the HK-2 cells was detected by real-time fluorescent quantitative PCR assay.Results:hUAT mRNA could be detected in all the samples.The level of hUAT mRNA in the control group was significantly lower than those in the other groups.The expression of hUAT gene was up regulated with the increase of uric acid concentration.The expression level of hUAT in group U3 was 116% of that in group C1.Conclusion:Uric acid can up-regulate the expression of hUAT gene.
