嘌呤核苷酸对慢性海洛因处理大鼠条件性位置偏爱与急性戒断的影响
Effect of purine nucleotides on conditioned place preference and acute withdrawal in chronic heroin dependent rats
【摘要】 目的研究嘌呤核苷酸对海洛因依赖大鼠的条件性位置偏爱与急性戒断的影响。方法将50只雄性Wistar大鼠随机分为对照组(腹腔注射生理盐水9d)、海洛因组(腹腔注射海洛因9d)、海洛因+单磷酸腺苷(AMP)+单磷酸鸟苷(GMP)组(腹腔注射海洛因及AMP及GMP9d)、海洛因+AMP组(腹腔注射海洛因及AMP9d)、海洛因+GMP组(腹腔注射海洛因及GMP9d)。于第0、5、9天测评各组大鼠条件性位置偏爱,即观察黑白箱内大鼠在位置偏爱实验箱白侧(伴药侧)的停留时间。通过纳洛酮催促,对大鼠急性戒断症状进行观察与评定。结果盐水对照组注射5d后在伴药侧停留时间明显缩短,注射9d后进一步缩短,对伴药侧产生条件性位置厌恶;海洛因组大鼠给药9d后在伴药侧停留时间显著延长,产生了对伴药侧的位置偏爱;海洛因+AMP+GMP组、海洛因+AMP组、海洛因+GMP组给药后在伴药侧停留时间无明显延长,说明补偿嘌呤核苷酸可抑制海洛因的位置偏爱效应。海洛因与嘌呤核苷酸同时作用的大鼠戒断后,上睑下垂、齿颤和咀嚼发生次数比海洛因单独作用组明显减少(P<0.05)。结论系统应用嘌呤核苷酸可抑制海洛因依赖大鼠精神依赖,还可减轻其部分急性戒断症状。
【Abstract】 Objective To determine the effect of purine nucleotides on conditioned place preference and acute withdrawal in chronic heroin dependent rats.Methods 50 male Wistar rats were randomly divided into control,heroin,heroin+AMP+GMP,heroin+AMP and heroin+GMP groups.Conditioned place preference(CPP) trial was done at 0,5,9 d.Naloxone was used to observe and evaluate the precipitated syndrome.Results In the CPP trial,the rats of control group showed obvious conditioned place aversion(CPA) because their total time staying in the drug-pairing side was decreased dramatically after 5 d treatment and dropped even more after 9 d treatment.In contrast,total time staying in the heroin-pairing side was increased significantly after 9 d heroin administration which proved that the rats formed CPP to heroin.The total time staying in the drug-pairing side of the heroin+AMP+GMP group,heroin+AMP group and heroin+GMP group were changed little which confirmed that purine nucleotides compensated with heroin inhibits rats’ CPP to heroin after 9 d heroin treatment.In acute withdrawal evaluation,the counts of ptosis,tooth chattering and chewing of heroin+AMP+GMP,heroin+AMP and heroin+GMP groups were significantly higher than those in heroin group(P<0.05).Conclusions Purine nucleotides can inhibit development of psychological dependence to heroin and alleviate some of the withdrawal symptoms.
