人参皂苷Rg1对AD模型大鼠脑片P-Tau、PKA影响的实验研究
The effects of Gensenoside Rg1 on the expressions of P-Tau and PKA on the brain slices of AD model rats
【摘要】 目的研究人参皂苷Rg1对AD模型大鼠脑片磷酸化微管相关蛋白(P-Tau)、蛋白激酶A(PKA)表达的调控作用。方法采用冈田酸诱导大鼠培养脑片Tau蛋白过度磷酸化,制备成AD模型,运用免疫组织化学、图像分析技术等方法,观察人参皂苷Rg1对各组大鼠脑片P-Tau、PKA表达的影响。结果模型组P-Tau、PKA表达水平明显高于空白对照组(P<0.01);人参皂苷Rg1各剂量组P-Tau、PKA表达水平虽高于空白对照组,但低于模型组(P<0.01或P<0.05),且大、中剂量组优于小剂量组;人参皂苷Rg1呈一定的剂量依赖性下调P-Tau、PKA的表达。结论人参皂苷Rg1能通过降低P-Tau水平、下调PKA的表达从而减缓神经纤维缠结、老年斑的形成,以发挥抗痴呆的作用。
【Abstract】 Objective To observe the effects of ginsenoside Rg1 on the expressions of P-Tau and PKA on the brain slices of AD model rats.Methods AD models were prepared by culturing rat brain slices with okadaic acid to induce Tau protein hyperphosphorylation.The effects of ginsenoside Rg1 on the expressions of P-Tau and PKA in AD model rat brain slices in each group were observed by immunohistochemistry and image analysis technology.Results The expressions of P-Tau and PKA in model group were significantly higher than those in normal control group(P<0.01).The expressions of P-Tau and PKA in Ginsenoside Rg1 groups were significantly lower than those in normal control group,but were higher than those in model group(P<0.01 or P<0.05),and Ginsenoside Rg1 showed a dose-dependent reduction P-Tau,PKA expression.Conclusions Ginsenoside Rg1 plays the role of anti-dementia through inhibiting the expressions of P-Tau and PKA and slowing the formation of neurofibrillary tangles.
