超声介导微泡造影剂对人脐静脉血管内皮细胞膜作用的研究
Research on the effect of ultrasound-mediated microbubbles contrast agent on HUVEC membrane
【摘要】 目的探讨超声介导微泡造影剂(MBCA)对人脐静脉血管内皮细胞(HUVEC)膜的作用。方法向两皿HUVEC中加入MBCA(商品名:SonoVue),用超声辐照,辐照后立即取一皿细胞送电镜室进行SEMS观察。另一皿细胞继续培养24h后进行SEMS观察。结果超声介导SonoVue可使HUVEC细胞膜的形态和结构发生明显异常改变。约23%细胞的细胞膜可见大小不等、数量不一的小孔。细胞继续培养24h后再进行SEMS观察,发现细胞膜的形态未见小孔样结构改变。结论超声介导SonoVue可使HUVEC细胞膜产生可逆性的小孔。
【Abstract】 Objective To research the effect of ultrasound-mediated microbubbles contrast agent (MBCA)-SonoVue on Human umbilical vein endothelial cells (HUVECs). Methods HUVECs were all treated with SonoVue. After ultrasound irradiation, one dish of HUVECs were observed by electron microscope, the others were observed after being cultured for 24 hours. Results Ultrasound-mediated SonoVue caused obviously abnormal alteration on the shape and structure of HUVEC membrane. Pores of different diameters and quantity appeared in the membrane of about 23% cell. After being cultured for another 24 hours, pore structure of the cells disappeared. Conclusions Ultrasound-mediated SonoVue causes reversible pores in HUVEC membrane.
