大鼠羊膜上皮细胞神经营养因子基因的表达
The differences of BDNF and NT-3 gene expression of the amnion epithelial cells (AECs) from pregnancy rats
【摘要】 目的建立胚胎不同时期羊膜上皮细胞(AECs)表达脑源性神经营养因子(BDNF)与神经营养素-3(NT-3)mRNA的时相图,找出AECs分泌BDNF、NT-3的最佳时期。方法采用实时荧光定量PCR检测胚胎不同时期(11,13,17,21d)AECs中BDNF与NT-3 mRNA的表达差异。结果绘制出大鼠AECs中BDNF、NT-3的基因表达差异时程图:BDNF的表达随着胚胎发育持续上调,至17d达到最高,然后其表达开始下调;NT-3的表达在13d达到最高,然后开始下调。结论初步确定13~17d是大鼠AECs分泌BDNF、NT-3的最佳时期。
【Abstract】 Objective To investigate the differences of BDNF and NT-3 gene expression of the amnion epithelial cells (AECs) from pregnancy rats and establish the phase diagram of gene expression to find the best time of BDNF and NT-3 secretion. Methods The differences of BDNF and NT-3 gene expression of the amnion epithelial cells (AECs) from different phase of pregnancy rats (E11, 13, 17 and 21 days) were detected by real time PCR. Results BDNF mRNA was up-regulated and lasted with embryonic development, reached the peak on E17 d, and then was down-regulated. NT-3 mRNA reached the peak on E13 d, and then was down-regulated.Conclusions The best time of BDNF and NT-3 secretion of AECs are from E13 d to E17 d.
