通心络超微粉对大鼠心肌梗死后心室重塑及其病理和心肌线粒体超微结构的影响

【摘要】 目的探讨通心络超微粉对大鼠心肌梗死后心室重塑(LVRM)及其病理和心肌线粒体超微结构的影响。方法130只SD大鼠随机取14只分为假手术组(0.9%NaCl灌胃,n=14,至实验终点时n=12)4w后处死。116只SD大鼠结扎冠状动脉前降支,术后存活的38只大鼠随机分为:急性心肌梗肆(AMI)模型组(0.9%NaCl灌胃,n=12)和通心络治疗组(1.0g.kg-1.d-1,n=13),卡托普利治疗组(15mg.kg-1.d-1,n=13)。心脏标本固定和病理分析,HE和Masson染色评价形态学特点。对术后2d和4w心脏超声数据分别进行分析。药物治疗4w后,用电子显微镜观察心肌细胞线粒体的变化。结果模型组和两治疗组间心肌梗死面积无明显差异,且三组大鼠术后2d和假手术组比较,左室舒张末期内径(LVEDD)明显增大(均P<0.001),而射血分数(EF)和短轴缩短率(FS)明显缩小(均P<0.001)。药物治疗4w后与模型组相比,两治疗组EF和FS明显增大(P<0.001),而左室舒张末期内径(LVEDD)明显缩小(P<0.001)。与术后2d相比,4w后模型组LVEDD明显增大(P<0.001),而两治疗组LVEDD明显减小(P<0.001)和EF和FS明显增加(P<0.001)。结论通心络超微粉可预防大鼠AMI后LVRM和改善心功能,这些作用基于通心络超微粉抗氧化应激作用。

【Abstract】 Objective To investigate the effects of Tongxinluo supermicropowder on left ventricular remodeling(LVRM) and the chances of myocardium pathology and mitochondria super-microcosmic structure in the rats with acute myocardial infarction(AMI). Methods 14 SD rats from 130 SD rats were randomly selected into sham operation group (fed with 0.9%NaCl, n=14, after 4 weeks, n=12), and were killed after 4 weeks. 116 SD rats underwent the left descending coronary artery ligation and 38 surviving rats were randomly assigned to AMI model group(fed with 0.9%NaCl, n=12),Tongxinluo supermicropowder treatment group (fed with Tongxinluo supermicropowder 1.0 g·kg-1·d-1, n=13) and captopril treatment group (fed with captopril 15 mg·kg-1·d-1, n=13). Morphological characteristics of rats′ myocardium were evaluated with HE, Masson stainning.The respective data of echocardiogram after 2 days and 4 weeks of surgery were analyzed. The changes of per-microcosmic structure of myocardial cell mitochondria were observed with electronic microscope after 4 weeks of treatment. Results There were no significant differences among model group and two treatment groups in infarction area. Compared with sham operation group after 2 days of surgery, the left ventricular end diastolic diameter (LVEDD) in model group and the two treatment groups were all significantly increased (all P<0.001), while the ejection fraction (EF) and shortening fraction (FS) were significantly decreased(all P<0.001). After 4 weeks of therapy, compared with model group, the EF and FS were significantly increased (P<0.001) while LVEDD were significantly decreased(P<0.001) in two treatment groups. Compared with after 2 days of surgery, LVEDD was significantly increased (P<0.001) in model group after 4 weeks of surgery, while LVED were significantly decreased (P<0.001) in two treatment group and the EF and FS were significantly increased (P<0.001) in two treatment groups. Conclusions Tongxinluo supermicropowder can prevent LVRM and improve cardiac function after AMI, which base on its antioxidation and anti-stress effects.
