电针促进帕金森模型小鼠黑质致密部蛋白激酶A、cAMP反应元件结合蛋白1表达

【摘要】 目的探讨电针能否促进帕金森病(PD)模型小鼠黑质致密部蛋白激酶A(PKA)、cAMP反应元件结合蛋白1(CREB1)的表达。方法以腹腔注射(30mg/kg)甲基苯基四氢吡啶(MPTP)诱导形成PD小鼠模型,电针"合谷"、"太冲"穴,频率2~100Hz,电压2~4V,疏密波,每日1次,每次20min,7次为1疗程,共治疗3个疗程后,采用免疫组化检测黑质致密部PKA、CREB1表达。结果模型组PKA、CREB1积分光密度低于空白组(P<0.05);电针组积分光密度高于模型组(P<0.01)。结论电针可促进PD模型小鼠黑质致密部PKA、CREB1表达。

【Abstract】 Objective To determine whether electroacupuncture (EA) treatment increase the expression of protein kinase A (PKA) and cAMP response element binding protein (CREB1)in substantia nigra (SNC) of Parkinson’s disease (PD) mice. Methods 24 male mice (C57BL/6J) were averagely and randomly divided into control,control EA,model and EA groups. PD model mice were induced by i.p. 1-methyl,4-phenyl-1,2,3,6 tetrahydropyridine. The mice were treated by EA with frequency from 2 to 100 Hz,volt arrange from 2 to 4 V,pointed on Hegu (LI4) and Taichong (LV3) every day. After 21 times treated,the mice were killed to detect the expression of PKA and CREB1 by immunohistochemistry. Results The OD value of PKA and CREB1 were decreased in model group than those in control group (P<0.05),and those in EA group were increased than those in model group(P<0.01).Conclusions EA therapy could increase the expression of PKA and CREB1 in PD mice SNC.
