丹参酮ⅡA对压力超负荷性心肌肥厚大鼠血液SOD、NO及Hyp含量的影响

【摘要】 目的:观察丹参酮ⅡA对压力超负荷性心肌肥厚大鼠血液超氧化物歧化酶(SOD)、一氧化氮(NO)、羟脯氨酸(Hyp)含量的影响。方法:健康成年SD雄性大鼠40只,随机分为假手术组、模型组、纈沙坦组(10 mg.kg-1)、丹参酮1组(5 mg.kg-1)、丹参酮2组(20 mg.kg-1)各8只,除假手术组外,其他组均采用胸主动脉部分缩窄术复制大鼠心肌肥厚模型。测量各组大鼠全心重量指数(HW/BW)、左心室重量指数(LVW/BW,即LVI);检测血清SOD活力、NO及Hyp含量。结果:模型组大鼠的HW/BW、LVI、Hyp含量明显高于假手术组,而SOD活力、NO含量明显低于假手术组(均P<0.05)。与模型组比较,丹参酮2组和纈沙坦组HW/BW、LVI及Hyp含量显著降低,SOD活力、NO含量显著升高;丹参酮1组与模型组各指标间差异无显著性意义。结论:丹参酮ⅡA对大鼠心肌肥厚有一定的保护作用,其机制可能与增加NO生成、抑制胶原合成、调节心脏局部氧自由基代谢等方面的作用有关,但<5 mg.kg-1的丹参酮ⅡA对心肌作用不明显。

【Abstract】 Objective: To investigate the effects of Tanshinone ⅡA(TSN) on blood superoxide dismutase(SOD),nitric oxide(NO),and hydroxyproline(HYP) in rats with pressure over-loading myocardial hypertrophy.Methods: Pressure over-loading myocardial hypertrophy model was established by thoracic aorta constriction operation.Forty healthy adult SD rats were randomly divided into the sham-operated group,the model group,low dose TSN group(5 mg/kg,intraperitoneal injection,i.p.),high dose TSN(20 mg/kg,i.p.),Valsartan group(10 mg/kg,gastrogavage).The cardiac index,and left ventricular weight index were measured,and the activity of SOD,and contents of NO and HYP were detected.Results: Compared with sham-operated group,cardiac index,left ventricular weight index,and content of HYP were increased,while content of NO and activity of SOD were decreased in the model group.Compared with model group,high dose TSN decreased cardiac index,left ventricular weight index,content of HYP,but increased the activity of SOD and the content of NO remarkably.There was no significant difference between low dose TSN and the model groups.Conclusion: TSN has protective effects on pressure over-loading myocardial hypertrophy of rats,and its mechanisms may involve in increasing production of NO,inhibiting production of collagen,and improving metabolism of oxygen-derived free radicals.
