【摘要】 目的:观察益肾强心合剂对慢性心衰大鼠心肌细胞凋亡的影响。方法:以皮下大量注射异丙肾上腺素(ISO)的方法制备慢性心衰大鼠模型。造模成功后随机分为模型对照组、卡托普利组及益肾强心合剂大、小剂量组,另设正常对照组,给药四周。TUNEL法检测心肌细胞凋亡指数,免疫组化方法检测心肌细胞Bax、Bcl-2蛋白表达,计算Bcl-2/Bax比值。结果:模型对照组心肌细胞凋亡指数、Bax促凋亡蛋白表达明显增加,Bcl-2抑凋亡蛋白表达减少、Bcl-2/Bax的比值明显降低,与正常对照组比较有显著性差异(P<0.05);益肾强心合剂大、小剂量组凋亡指数、Bax蛋白显著降低,Bcl-2蛋白明显增加、Bcl-2/Bax比值明显升高,与模型对照组比较有显著性差异(P<0.05)。结论:益肾强心合剂对慢性心衰心肌细胞凋亡有良好的抑制作用,是防治慢性心衰(CHF)的有效药物。更多还原

【Abstract】 Objective:To study the effects of Yishenqiangxin mixture on cardiac myocyte apoptosis of chronic congestive heart failure for rat model.Methods:The models of heart failure in the study was induced by subcutaneously injecting a large dose of ISO and were randomly divided into model group,captopril group,high dosage group of Yishenqiangxin mixture,low dosage group of Yishenqiangxin mixture and in addition to setting up normal group.The drugs were taken by oral for 4 weeks.Cardiac myocyte apoptosis were detected with TUNEL method and the apoptosis index was calculated.The expressions of Bax、Bcl-2 were detected with immunohistochemical method and the ratio of Bcl/Bax was calculated.Results:Compared with that of normal group,myocardial apoptosis index increased significantly,expression of Bax increased significantly but expression of Bcl-2 and the Bcl-2 to Bax ratio decreased significantly(P<0.05,P<0.01).Compared with that of model group,myocardial apoptosis index decreased significantly in high and low dosages of Yishenqiangxin mixture(P<0.05),expression of Bax decreased but expression of Bcl-2 and the Bcl-2 to Bax ratio increased significantly in the same group(P<0.05).Conclusions:Yishenqiangxin mixture can inhibit cardiac myocyte apoptosis of chronic congestive heart and Yishenqiangxin mixture is an effective drug in treating CHF.更多还原
