三氯苯达唑对斯氏狸殖吸虫童虫作用的光镜和透射电镜观察
Light microscope and ultrastructural changes in juvenile Pagumogonimus skrjabini treated by triclabendazole
【摘要】 目的探索三氯苯达唑治疗大鼠斯氏狸殖吸虫童虫感染的最适剂量;进一步探讨三氯苯达唑杀灭斯氏狸殖吸虫的机理。方法60只SD大鼠随机分成实验组和对照组,每组15只。将斯氏狸殖吸虫囊蚴经腹腔注射感染各组大鼠。感染后第39 d分别用不同剂量三氯苯达唑[50 mg/(kg.d)×4 d、100 mg/(kg.d)×4 d2、00 mg/(kg.d)×4 d]治疗实验组大鼠,治疗结束后第1、15、22 d剖杀大鼠,每组每次剖杀5只,检获的虫体分别用光学显微镜和透射电镜观察其组织结构的改变。结果三氯苯达唑对童虫体壁和肠壁的组织结构有不同程度的损坏,但低剂量组轻微,中剂量组对虫体的破坏作用比低剂量组强,但尚不致虫体死亡,而高剂量组对虫体的破坏是致死性的。对照组大鼠检获的虫体未出现病理改变。结论三氯苯达唑治疗大鼠斯氏狸殖吸虫童虫感染的最适剂量约为200 mg/(kg.d)。三氯苯达唑对斯氏狸殖吸虫的作用机理除了损伤虫体体壁外,还可破坏虫体肠管组织,导致寄生虫的消化、吸收、分泌、排泄等生理功能和生化代谢障碍而杀伤或杀灭虫体。
【Abstract】 Objective To determine the best dosage of triclabendazole in rats infected with the juvenile worms of Pagumogonimus skrjabini and to explore the mechanism by which triclabendazole kills P.skrjabini.Methods Sixty rats were randomly divided into four groups(three experimental groups and a control group) with 15 rats in each group.The 60 rats were eacHⅠnjected in the abdominal cavity with juvenile P.skrjabini.Rats of the experimental groups were treated with different doses [50 mg/(kg·d)× 4 d,100 mg/(kg·d)× 4 d,and 200 mg/(kg·d)× 4 d] of triclabendazole on the thirty-ninth day after infection.On the first,fifteenth and twenty-second days after finishing the treatment,five rats in each group at each time point were dissected.The structural changes of the worm tissues were observed under both a light microscope and a transmission electron microscope.Results Triclabendazole had different extents of breakage in the ultrastructure of the body wall and intestinal wall of juvenile of P.skrjabini.The effect of the low-dose on worms was less,the destructive effect of the middle-dose on worms was stronger than the low-dose but was not deadly,and the destructive effect of the high-dose on worms was deadly.Worms recovered from rats in the control group showed no pathological changes.Conclusion The best dose of triclabendazole to treat rats infected with the juvenile worms of P.skrjabini is about 200 mg/kg·d.The mechanism of triclabendazole killing of P.skrjabini is damage to the tissue of both the body walls and intestines,which result in handicapping of both physiological function and biochemical metabolism of assimilation,absorb,exudation,excretion,etc.of the parasite.The worms are ultimately killed or wounded.
