蝮蛇蛇毒蛋白指纹图谱研究及应用
Fingerprints of Venom from Agkistrodon halys Pallas. and Its Application
【摘要】 采用高效毛细管电泳法,研究了江浙蝮蛇蛇毒蛋白指纹图谱。以10mmol/L硼砂(pH9.1,含4.62%正丁醇、3.5%十二烷基磺酸钠和0.64%正辛烷)为电泳缓冲液,紫外检测波长195nm,电压10kV。纤溶酶峰为参照物峰,确定了15个共有峰,并分析比较了11种不同来源的江浙蝮蛇蛇毒蛋白。结果显示,指纹图谱中主要峰群的整体图貌基本一致,但各成分含量的对比值有所不同。
【Abstract】 The capillary electrophoresis fingerprints was established to evaluate the quality of venom from Agkistrodon halys Pallas.,at the detection wavelength of 195 nm,applied voltage of 10 kV. The background electrolyte consisted of 10 mmol/L sodium borate solution (pH 9.1,containing 4.62% 1-butanol,3.5% SDS,and 0.64% octane). Fifteen co-possessing peaks were selected as the fingerprints and plasmin was selected as the reference peak. By analyzing and comparison to 11 batches of venom,it was showed that the chromatograms were similar while the ratio of common peak area of different samples was different.
