福州地区绝经后妇女骨钙素基因Hind Ⅲ多态性与骨密度的关系

【摘要】 目的探讨绝经后妇女骨钙素基因(OC)HindⅢ多态性与骨密度的关系。方法用双能X线骨密度仪检测247例绝经后妇女的腰椎、股骨颈、大转子和Ward s三角骨密度,应用PCR-RFLP技术检测OC基因HindⅢ多态性。结果①OC基因型分布频率为HH型4.9%,hh型49%,Hh型46.1%。等位基因频率为H27.94%,h72.06%,基因型分布符合Hardy-Weinberg定律。②HH基因型与hh型在股骨颈、大转子、Ward s三角区骨密度有显著性差异(P<0.05);HH基因型与Hh型在大转子、Ward s三角区骨密度也有显著性差异(P<0.05)。③有h等位基因的股骨颈、大转子、Ward s三角区骨密度显著低于无h等位基因(P<0.05)。结论福州地区绝经后妇女OC基因多态性与骨密度存在相关性,有h等位基因绝经后妇女在股骨颈、大转子、Ward s三角区有低骨密度风险。

【Abstract】 Objective To investigate the association between Hind Ⅲ polymorphism of OC gene with bone mineral density(BMD) in postmenopausal women.Methods The BMD of lumbar spine,femoral neck,trochanter and Ward’s triangle of 247 postmenopausal women were measured by dual energy X-ray absorptiometry.The genotype of OC gene was determined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).Results ①The distribution of OC genotypes were HH genotype 4.9%,hh 49%,Hh 46.1%,respectively.The H allelic gene frequencies reached 27.94%,while h was 72.06%.The distribution followed the Hardy-Weinberg equilibrium.② The difference was found in femoral neck、trochanter and Ward’s triangle BMD between HH and hh genotypes(P<0.05);there is a significant difference in BMD of trochanter and Ward’s triangle between HH and Hh genotype(P<0.05).③The postmenopausal women of presence h had lower BMD than those absence h in femoral neck,trochanter and Ward’s triangle,the difference reached statistical significance.Conclusion Between genotype of OC gene and BMD of the postmenopausal women have significant association,the presence(h) in the postmenopausal women is related to low BMD in femoral neck,trochanter and Ward’s triangle.
