TRAMP前列腺癌小鼠模型肿瘤的病理特点

【摘要】 目的:研究TRAMP前列腺癌小鼠模型肿瘤的病理特点,评估其在前列腺癌研究中的价值。方法:10只28周龄TRAMP小鼠,提取鼠尾DNA作基因型鉴定,安死后,探查腹、盆腔淋巴结、肝脏、肺等器官,观察有无转移灶,摘取前列腺,10%福尔马林固定过夜后转移至70%酒精备用。固定组织石蜡包埋,5um厚度切片后,HE染色作组织学检查,T抗原行免疫组化染色。结果:共筛查98个子代,雌性子代T抗原PCR阳性率为50%,雄性子代T抗原阳性率为48.5%。28周龄TRAMP小鼠前列腺肿瘤成瘤率为100%(10/10)。前列腺病变表现为从早期前列腺上皮内瘤到晚期腺癌、转移癌演变的各种特点。10例中有6例出现腹盆腔淋巴结肿大,3例出现肺部转移,1例出现肝脏转移,1例出现上肢皮下组织转移。免疫组化染色见前列腺肿瘤组织中高表达T抗原。结论:TRAMP小鼠模型的病理特点适合前列腺癌发病机制及其进展研究。

【Abstract】 Objective:To evaluate the pathological characteristics of prostate cancer in a transgenic animal model. Methods:At the time of sacrifice,peritoneal lymph nodes,liver and lung from ten 28-week-old Transgenic Adenocarcinoma of the Mouse Prostate (TRAMP) mice were checked for macroscopic evidence of metastasis. The prostate tissues were taken and fixed overnight in 10% neutral buffered formalin,and then transferred to 70% ethanol before standard tissue processing. Fixed tissues were embedded in paraffin,and sections of 5 μm were mounted on slides. Sections were routinely stained with hematoxylin and eosin (HE) for pathological study. T antigen immunohistochemical staining was also performed. Results:In the total 98 off-springs,the positive expression of T antigen was 50% in the female and 48.5% in the male. At the age of 28 weeks,100% TRAMP mice displayed primary tumors or metastasis. TRAMP model showed spontaneous progressive stages of prostatic disease with time from early lesions of prostatic intraepithelial neoplasia (PIN) to local adenocarcinoma and finally to metastatic disease. Six out of ten TRAMP mice had metastases in peritoneal lymph nodes. The incidence of lung metastasis,liver metastasis and subcutaneous tissue metastasis was 3/10,1/10,and 1/10,respectively. Immunohistochemistry of T antigen in prostate tumors showed strong positive expression. Conclusion:TRAMP is a characterized animal model for the study of pathogenesis and progression of prostate cancer.
