龟鹿二仙胶汤含药血清对体外软骨细胞表型BMSCs凋亡的影响

【摘要】 目的:观察龟鹿二仙胶汤含药血清干预软骨细胞表型化骨髓质基质干细胞(BMSCs)凋亡的影响。方法:抽取兔骨髓液,经体外分离、纯化、培养获得免BMSCs,将龟鹿二仙胶汤含药血清诱导出的软骨细胞表型化BMSCs,用IL-1β诱导凋亡,分别于24h、48h、72h后,采用透射电子显微镜、免疫荧光组织化学、流式细胞仪等实验技术,研究龟鹿二仙胶汤对软骨表型化BMSCs凋亡的影响。结果:龟鹿二仙胶汤能减少软骨表型化BMSCs凋亡的数量及程度,降低细胞凋亡率。结论:龟鹿二仙胶汤可以明显抑制或逆转软骨表型化BMSCs凋亡,能显著延缓软骨表型化BMSCs的凋亡。

【Abstract】 Objective:To observe the effect of Guilu Erxian decoction(GED) serum on the apoptosis of chondrocyte phenotype differentiation of bone marrow-derived mesenchymal cells(BMSCs).Methods:The BMSCs were isolated from rabbit bone marrow,and the ceils were extracted,purified and cultured in vitro.The cells that show chondrogenic phenotype differentiation of BMSCs were induced into apoptosis cells by IL-1β.after 24,48 and 72 hours,transmission electron micro scope,immune histochemistry,flow cytometry analysis were used to observe the effect on chondrocyte phenotype of BMSCs apoptosis.Results:GED decreased quantity and degree of apoptosis in Chondrogenic Phenotype cells and reduced the apoptosis of BMSCs.Conclusion:GED can significantly prevent or reverse the apoptosis of chondrogenic phenotype and de lay the apoptosis of chondrogenic phenotype.
