甲氨蝶呤聚乳酸或聚乙交酯丙交酯纳米粒的制备及其特性
Preparation and properties of methotrexate-loaded PLA or PLGA nanoparticles
【摘要】 目的:制备甲氨蝶呤聚乳酸(PLA)或聚乙交酯丙交酯(PLGA)纳米粒并研究其性质。方法:以生物相容性好的PLA或PLGA为载体,采用溶剂挥发法发展了一类新的甲氨蝶呤(MTX)纳米制剂的方法。结果:PLA或不同的单体比(L/G,共聚物中乳酸与乙醇酸的质量比)的PLGA的MTX制剂的平均粒径在180~210nm之间,多分散系数在0.300附近,Zeta电位小于-18.0mV;通过该方法制备的MTX纳米制剂的包封率高,具有明显的缓释作用。结论:本研究为开发MTX新型纳米制剂提供了实验依据。
【Abstract】 OBJECTIVE To Prepare methotrexate(MTX)-loaded polylactic acid(PLA) or poly(lactic-co-glycolic acid)(PLGA) nanoparticles and study nanoparticles properties.METHODS MTX-loaded PLA or PLGA nanoparticles were prepared by emulsion solvent evaporation method with good biocompatible PLA or PLGA as the carriers.RESULTS Mean size of the MTX nano-pharmaceutics of PLA or PLGA with different monomer ratio(in copolymer: L/G ratio=m(lactide)/m(glycolide)) was 180-210 nm,polydispersity index was about 0.300,and Zeta potential was less than-18.0 mV.The MTX pharmaceutics had high entrapment efficiency,and showed evidently sustained release effect.CONCLUSION This study will provide an experiment basis for development of new kind of nano-pharmaceutics of MTX.
