肾康丸对糖尿病大鼠肾脏Ⅰ型胶原和转化生长因子-β1表达的影响
Renal protective effect of Shenkang pills on diabetic rats
【摘要】 目的:观察肾康丸对糖尿病(DM)大鼠模型的肾功能及Ⅰ型胶原合成的影响。方法:用链脲佐菌素(STZ)腹腔注射法诱导建立大鼠DM模型,模型成功后随机分为模型对照组、开搏通组、肾康丸组3组,另设正常对照组。治疗8周,观察治疗期间及治疗后大鼠一般状况、血糖、24h尿蛋白、肌酐、尿素氮、肾质量、相对肾质量,并用实时荧光定量PCR测定肾脏Ⅰ型胶原(collagenⅠ)和转化生长因子-β1(TGF-β1)mRNA的表达情况。结果:造模3组均出现DM及肾脏损害,肾脏collagenⅠ、TGF-β1mRNA的表达明显增加。肾康丸和开搏通均可改善大鼠基本状况、肾功能、尿蛋白,下调肾脏组织collagenⅠ、TGF-β1mRNA的表达;肾康丸组血糖浓度显著降低(P<0.01),其降血糖疗效优于开搏通(P<0.01)。结论:肾康丸可以减少col-lagenⅠ、TGF-β1的产生,延缓糖尿病肾病进程。
【Abstract】 OBJECTIVE To investigate the effect of Shenkang pills on renal function and collagenⅠsynthesis in diabetic rats.METHODS The diabetic rat modls were induced by intraperitoneal injection of streptozotocin(STZ) and randomly divided into model control group, Capoten group and Shenkangwan group. Some normal other rats were used as normal control group. All rats were treated with corresponding drugs for 8 weeks. During and after the treatment, the general state, blood glucose levels, excretion rate of the 24hour urine protein,serum creatinine and blood urea nitrogen contents, kidney weight and relative kidney weight were measured.The mRNA of collagenⅠand TGF-β1 in the kidney also were detected by quantitative real-time PCR.RESULTS Diabetes mellitus and renal lesions occurred in the three model groups. The expression of collagenⅠand TGF-β1 mRNA of the kidney in diabetic rats increased obviously. Shenkang pills could improve the general state and renal function of the diabetic rats, decrease the blood glucose levels and the excretion rate of the 24 hour urine protein, reduce the expression of collagenⅠand TGF-β1 mRNA in kidney.CONCLUSION Shenkang pills suppressed synthesis of collagenⅠand TGF-β1. Shenkang pills could ameliorates renal lesions in diabetic rats.
