阳离子聚乙烯亚胺高分子聚合物对青霉素类抗生素的抑菌增效作用
Influence of Polyethyleneimine on in Vitro Bactericidal Activity of Penicillin
【摘要】 目的评价聚乙烯亚胺(polyethylenimine,PEI)多阳离子聚合体对青霉素类抗生素体外抑制铜绿假单胞菌(Pscudomonas aeruginosa,100609)活性的影响。方法采用光密度法测定PEI多阳离子聚合体分别与4种青霉素类抗生素联合使用对铜绿假单胞菌的最小抑菌浓度的影响。同时使用光密度法测定不同浓度相对分子质量为10000PEI多阳离子聚合体与亚最小抑菌浓度(11.5%MIC的浓度)的替卡西林共同作用对P.aeruginosa,100609的抑菌作用并计算抑菌百分数。结果相对分子质量为10000,260mg·L-1的PEI分别使替卡西林,羧苄西林,氨苄西林和哌拉西林的MIC降低4~64倍。在细菌动力学致死-增殖研究中,发现PEI可显著增强替卡西林对P.aeruginosa,100609的抑菌作用,且其作用与PEI的浓度存在正相关性,表明PEI与青霉素类抗生素具有协同作用。结论PEI多阳离子聚合体可以显著增强青霉素类药物对P.aeruginosa,100609的抑菌活性。
【Abstract】 OBJECTIVE To evaluate the influence of a synthetic polycationic polymer,polyethylenimine(PEI),on the in vitro bactericidal activity of penicillins antibiotics.METHODS The minimal inhibitory concentration(MIC) and time-killing rate of different penicillins alone and in combination with PEI against a resistant clinical isolate,P.aeruginosa 100609 were investigated by the optical densitometric method.The effect of bacteriostasis by different concentrations of the molecular weight of 10 000 PEI and ticarcillin(11.5% MIC concentrations) combined against P.aeruginosa were determined and bacteriostasis percentage was calculate.RESULTS MICs of ticarcilline,carbenicilline,ampicillin and piperacillin against,P.aeruginosa were efficiently decreased by 4-64 times with 260 mg·L-1,10 000 of PEI respectively.In a kinetic bacteria killing-regrowth study,a combination of ticarcillin(at concentration of 11.5% MIC) and PEI(260 mg·L-1,10 000) significantly reduced the viable bacterial count compared to either ticarcillin or PEI alone enhanced bactericidal activity of penicillins with PEI was synergistic and dependented on the concentration of PEI.CONCLUSION The PEI could dramatically increase bactericidal activity of penicillins antibiotics against a resistant Pseudomonas aeruginosa.
