结直肠癌组织的二维荧光差异凝胶电泳分析
Analysis of Colorectal Cancer by Two-dimensional Differential Gel Electrophoresis
【摘要】 目的以二维荧光差异凝胶电泳(2-D DIGE)技术分析结直肠癌(CRC)组织及正常结直肠组织的蛋白质表达差异。建立二者间的蛋白质表达差异图谱。方法6对样品分别取自6例CRC手术切除的新鲜的癌和正常肠黏膜组织,进行2-D DIGE,TyphoonTM多功能扫描仪进行图像扫描,并以DeCyderTM2-D差异分析软件分析。结果正常肠黏膜组织和癌组织有统计学差异(P<0.05)的表达蛋白点共有315个,癌组织中表达上调(>1.5倍)的有138个,下调(>1.5倍)的有103个。结论建立了CRC与正常肠黏膜组织的2-D DIGE图谱,发现并经统计学证实癌组织中有表达上调或下调的蛋白存在。
【Abstract】 Objective To analyze the differential protein expression between colorectal cancer (CRC)tissue and adjacent normal mucosa by two-dimensional differential gel electrophoresis(2-D DIGE)and establish the differential expression profiles of them. Methods 6 paired samples,tumor and adjacent normal tissue,were taken from fresh resections from 6 CRC patients. The samples were analyzed by 2-D DIGE, TyphoonTM multifunctional scanner and two-dimensional differential analysis software of DeCyderTM. Results There were 315 protein spotfeatures changing in abundance across the six patient samples with statistical variance of the normal:tumor spot volume ratio(P < 0.05),including 138 and 103 spot-upregulated and downregulated more than 1.5-fold,respectively. Conclusion The differential expression profiles of 2-D DIGE of CRC and adjacent normal mucosa tissues were established. Upregulated and downregulated proteins,which could be confirmed statistically,were found in CRC tissue comparing with normal mucosa.
