【摘要】 目的鉴定WNK4基因启动子结构,初步探讨WNK4基因表达的调节机制。方法应用5′RACE方法确定WNK4基因转录起始点;构建WNK4基因启动子报告基因载体,并应用ELISA法检测启动子活性;应用软件分析预测WNK4基因启动子区顺式作用元件。结果初步确定了WNK4基因转录起始位点;构建了WNK4基因启动子报告基因载体,并通过ELISA方法确定了转录起始点上游1 275 bp具有高转录活性,并且软件预测该区域内存在多个顺式作用元件,初步证明该区域为WNK4基因的启动子序列。结论鉴定了WNK4基因的启动子区,并对其功能进行了初步鉴定。更多还原

【Abstract】 Objective To identify the promoter of WNK4 gene and explore the regulatory mechanism of its expression.Methods 5′ RACE was used to identify the transcription start site of WNK4 gene.Then reporter vectors of WNK4 gene promoter were constructed,and ELISA was used to determine the promoter activity.The cis-acting elements within the WNK4 promoter were analyzed.Results The transcription start site of WNK4 gene was identified.The WNK4 promoter reporter vectors were constructed,and the results of ELISA showed high transcriptional activity of the 1 275 bp upstream of the transcription start site.A lot of cis-acting elements were identified within this region by software analysis,suggesting this region was the WNK4 gene promoter sequence.Conclusion The promoter region of the WNK4 gene is identified,and its function is analyzed primarily.更多还原
