氯吡格雷对支气管哮喘小鼠血小板源性P选择素及肺泡中性粒细胞的影响

【摘要】 目的观察氯吡格雷对支气管哮喘小鼠的P选择素及肺泡中的中性粒细胞的影响。方法雌性BALB/c小鼠共30只随机分成3组,正常对照组(对照组)、哮喘模型组(模型组)、氯吡格雷治疗组(治疗组)。用流式细胞仪测定血小板膜上P选择素的阳性率,并观察病理组织学及细胞学的相应变化。结果对照组、模型组和治疗组P选择素表达的阳性率分别为(1.23±1.02)%、(4.52±1.45)%、(1.42±1.12)%,模型组P选择素较对照组及治疗组增高(P<0.05);对照组、模型组及治疗组凝血酶诱导P选择素数值分别为(45.12±4.03)%、(44.68±5.34)%、(44.23±5.42)%,三者之间无显著性差异(P>0.05);对照组、哮喘组及治疗组肺泡灌洗液中性粒细胞分别为(6.64±1.86)%、(25.60±2.66)%、(7.23±2.02)%,模型组较正常对照组和治疗组增高(P<0.05)。结论支气管哮喘小鼠存在血小板的活化,氯吡格雷可以减轻支气管哮喘中的炎性反应。

【Abstract】 Objective To observe the effects of Clopidogrel on P-selectin derived from platelets and neutrophils in the asthmatic mice. Methods Female BALB/c mice were divided randomly into 3 groups:control group,asthmatic group and treated group by clopidogrel. The P-selectin positive rates of platelet were measured by flow cytometer,histopathologic and cytologic changes were studied as well. Results The P-selectin positive rates of platelets in control group,asthmatic group and treatment group were(1.23±1.02)%,(4.52±1.45)% and(1.42± 1.12)%,respectively. There was a significant difference in the P-selectin positive rates of platelets expressed in control group,asthmatic group and treatment group(P < 0.05). The P-selectin of platelets positive rates after activated by thrombin were not significant different in control group,asthmatic group and treatment group(P > 0.05). Their values were(45.12±4.03)%,(44.68±5.34)% and(44.23±5.42)%,respec-tively. The percentage of neutrophil of bronchial alveo lavage (BAL)in control group,asthmatic group and treatment group were (6.64 ± 1.86)%,(25.60±2.66)% and (7.23±2.02)%,respectively. The percentage of neutrophil of BAL in asthmatic group was significant higher than in control group or treatment group(P < 0.05). Conclusion Platelets activation was found in the asthmatic model mice whereas clopi-dogrel can reduce the inflammation during the asthmatic attack.
