【摘要】 目的了解HIV/AIDS感染者进行CD4+T淋巴细胞检测时血样经荧光染色后随着放置时间的延长,CD4+T淋巴细胞计数的变化。方法用FACSCount流式细胞仪对运城市HIV/AIDS病人送检的30份血样采集后放置8 h进行荧光染色,染色后再放置30 min、24 h、48 h、72 h、96 h后分别进行CD4+T淋巴细胞计数。并将结果进行平均值(x-)、标准差(s)和变异系数(CV)的统计计算。结果同一样品染色后放置30 min至96 h内检测的数据差异均<20%,为可接受范围。其中血样经荧光染色后再放置不同时间检测的30份血样中有21份五次重复测定值的CV值<5%,占70%。相关性分析显示:五个时间点上的检测结果相互之间都具有很好的相关性,且相关系数(r值)均≥0.99。结论在实验室其他条件(温度、试剂等)不变的情况下,用FACSCount流式细胞仪检测CD4+T淋巴细胞,血样经荧光微球染色后放置时间延长至96 h五个时间点的CV%值<20%,对检验结果影响不大。更多还原

【Abstract】 Objective To define the influence of prolonged stored time of blood samples after antibody staining on CD4+ T-cell counts.Methods Thirty blood samples were collected for this study.Antibody staining was conducted at 8 hours after blood collection.CD4+ T-cell counts were acquired at 30 minutes,24,48,72 and 96 hours after antibody staining.Mean,SD and CV values of CD4+ T-cell counts at different time points were calculated and compared.Results CV values for samples with different stored time were all <20%,among which,70%(21 out of 30 samples)were <5%.Correlation analysis revealed that CD4+ T-cell counts at different time points were highly correlated with each other.Conclusion Under the same experimental conditions,samples′ stored time has little effect on CD4+ T-cell counts determined by FACSCount flow cytometry.更多还原
