前列腺素E1对低硒高镉饲料饲养大鼠心室乳头肌细胞的电生理作用

【摘要】 目的:观察前列腺素E1对低硒高镉饲料饲养大鼠心室乳头肌细胞电生理的影响。方法:用细胞内微电极技术分别观察普通饲料对照、本实验构成饲料对照、单纯高镉、高镉高前列腺素E1、低硒高镉、低硒高镉伴高前列腺素E1、高硒高镉、高硒高镉伴高前列腺素E1饲料饲养14周后大鼠心肌细胞电生理指标静息电位(RP)、动作电位幅度(APA)、动作电位时程(APD50、APD90)的改变。结果:与2个对照组比较,高镉高前列腺素E1组、高硒高镉伴高前列腺素E1组各检测指标无变化;单纯高镉组RP、APA降低,APD50、APD90延长;低硒高镉组RP、APA降低,APD50、APD90明显延长;低硒高镉高前列腺素E1组、高硒高镉组均表现为仅RP降低,APA、APD与2个对照组比较差异无显著性意义。结论:低硒高镉可明显改变心肌细胞的电生理特性,前列腺素E1可以对抗此变化。

【Abstract】 OBJECTIVE:To observe the effects of prostaglandin E1(PE1)on the electrophysiological property of ventricular papillary muscle cells of rats fed with low selenium(Se)and high cadmium(Cd)diet.METHODS:Using intracellular mi-croelectrode technique,the changes of electrophysiological indexes of myocardial cells including resting potential(RP),action potential amplitude(APA),action potential duration(APD50 and APD90)in rats after uptake of different diet(common diet control,trial diet control,high Cd,high Cd+high PE1,low Se+high Cd,low Se+high Cd+high PE1,high Se+high Cd,or high Se+high Cd+high PE1)for 14 weeks were observed.RESULTS:Compared with two controls,there were no significant changes in electrophysiological indexes in high Cd+high PE1 group or high Se+high Cd+high PE1 group;however,in high Cd group,RP and APA decreased while APD50 and APD90 prolonged;in low Se+high Cd group,RP and APA decreased,APD50 and APD90 prolonged significantly;in low Se+high Cd+high PE1 group and in high Se+high Cd group,only RP decreased,but no significant differences were noted in APA and APD as compared with two controls.CONCLUSION:Low Se and high Cd could markedly change the electrophysiological properties of rats’ myocardial cells;however,PE1 may protect myocardial cells against this change.
