HPLC法测定虎杖中大黄素-8-O-β-D-葡萄糖苷的含量

【摘要】 目的:建立以高效液相色谱法测定虎杖中大黄素-8-O-β-D-葡萄糖苷含量的方法。方法:采用D101大孔吸附树脂对虎杖样品进行预处理;色谱柱为Kromasil C18(250mm×4.6mm,5μm),柱温为40℃,流动相为乙腈-水(40∶60),流速为1.0mL·min-1,检测波长为420nm。结果:大黄素-8-O-β-D-葡萄糖苷的检测浓度在5~200μg·mL-1范围内与峰面积积分值呈良好的线性关系;平均回收率为98.63%,RSD=0.99%(n=6)。结论:本方法简便、快速、准确,可用于虎杖药材的质量控制。

【Abstract】 OBJECTIVE:To determine the concentration of emodin-8-O-β-D-glucoside in Polygonum cuspidatumby HPLC. METHODS: The giant knotweed rhizome sample was pretreated by D101 macroporous adsorptive resins. The separation of sample was performed on Kromasil C18(250 mm×4.6 mm, 5μm) with column temperature set at 40 ℃. The mobile phase consisted of acetonitrile-water (40∶60) at a flow rate of 1.0 mL·min-1 and a detection wavelength of 420 nm. RESULTS: The linear range of emodin-8-O-β-D-glucoside was 5~200 μg·mL-1 with an average recovery of 98.63%(RSD=0.99%,n=6). CONCLUSION: The method applied in the study is simple, rapid, accurate, and suitable for the quality control of determination of emodin-8-O-β-D-glucoside in Polygonum cuspidatum.
