通络方药对糖尿病大鼠胰岛β细胞的保护作用
Protective effects of Tongluo recipe on damaged beta-cells of rat with streptozotocin-induced diabetes
【摘要】 目的研究通络方药(TLR)对链脲佐菌素(STZ)所致大鼠胰岛β细胞损伤的保护作用。方法建立糖尿病大鼠模型前8天开始给予TLR直至实验结束。实验结束时测血糖、体重;取胰腺测定胰腺匀浆中丙二醛(MDA)及谷胱甘肽过氧化物酶(GSH-Px)水平,透射电镜检测胰岛β细胞。结果与正常组相比,STZ组胰腺内MDA水平明显升高(P<0.05),GSH-Px活性明显降低(P<0.05),血糖明显升高。TLR预处理明显降低STZ组大鼠的血糖水平,同时降低大鼠胰腺匀浆内MDA含量(P<0.05),升高GSH-Px活性(P<0.05)。透射电镜显示TLR预处理使胰岛β细胞的损伤明显减轻。结论 TLR通过减轻胰腺内氧化应激水平保护胰岛β细胞完整并有效防止STZ诱导的糖尿病发生。
【Abstract】 Objective To investigate the protective effects of Tongluo Recipe (TLR) on damaged beta-cells of rat with streptozotocin(STZ)-induced diabetes. Methods TLR was given for 8 days prior to induction of diabetes with STZ until the end of the study (4 weeks). Blood glucose levels and body weights were examined, Malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) activity were measured in pancreatic homogenates at the end of the study. Pancreatic beta-cells were examined by electron microscope. Results STZ induced a significant increase in MDA levels (P<0.01) and a marked decrease in GSH-Px activity (P<0.05) in pancreatic tissue. Blood glucose levels were significantly increased in STZ-treated rats only. Pretreatment of STZ-tredated rats with TLR decreased the incidence of diabetes compared with STZ-treated rats without TLR. At the same time, markedly reduced MDA production (P<0.05) and elevated GSH-Px activity (P<0.01) were apparent in the TLR-pretreated rats. Transmission electron microscope observations showed that the STZ-induced beta cell damage was attenuated in rats pretreated with TLR. Conclusions TLR exerts its protective effect on hyperglycemia and beta-cell damage through reducing the oxidative stress and preserving pancreatic beta-cell integrity.
