大青叶中4(3H)喹唑酮提取工艺的研究

【摘要】 将4(3H)喹唑酮含量作为大青叶的质控指标,采用正交试验方法考察了药材粒度、提取溶剂、提取时间、溶剂倍量等因素对提取的影响。分别进行热回流提取、超声波提取和微波提取工艺研究,优选4(3H)喹唑酮的最佳提取工艺条件。结果表明,在提取因素方面,药材粒度是热回流提取的主要影响因素,溶剂倍量是微波提取的主要影响因素,乙醇浓度是超声波提取的主要影响因素。当以提取速率(提取率与提取时间的比值)为分析指标时,微波提取工艺要明显高于其他提取工艺,超声波提取工艺次之。三种提取方法中,热回流提取工艺的提取率最高,其最佳工艺条件为A2B3C1D1,即药材粒度0.30 mm、700 mL/L乙醇、热回流时间1 h、8倍量溶剂。

【Abstract】 The content of 4(3H)quinazolinone from Folium isatidis was used an index to conduct heat reflux,ultrasonic and microwave extractions for the optimal conditions of an orthogonal test in which factors were materials size,extractant concentration,extraction time and solvent volume on the extraction of 4(3H)quinazolinone.The results showed that the materials size,solvent volume and concentration of alcohol were the main influence factors in the heat reflux extraction,microwave extraction and ultrasonic extraction respectively.Microwave extraction was obviously higher than the two others and ultrasonic extraction took the second place when using extraction velocity as analysis index.Heat reflux extraction was the best of the three extraction technologies in extractional rate of 4(3H)quinazolinone.The optimal conditions of heat reflux extraction were A2B3C1D1,i.e.scatter medical materials were 0.30 mm in particle size along with 8 times of 700 mL/L alcohol for one hour.
