黄绿蜜环菌产纤溶酶的发酵条件优化研究

【摘要】 为了优化密环菌产纤溶酶的发酵条件,通过单因素试验和响应面试验设计,以麸皮为碳源,酵母膏为氮源,加入0.1%MgSO4,用豆汁配制培养基,pH4.0时得到单因素最大产酶效果。分别以2%麸皮、2%酵母膏、0.1%MgSO4为主要优化因子,选择适当步长,设计响应面试验,回归拟合试验数据,得到二次回归模型。该模型R2=0.8709,在90%的概率水平上显著。用该模型拟合得到的各因素的最适加入量分别为麸皮2.97%,酵母膏2.30%,MgSO40.147%,预测的最高产酶量达到410IU/mL。

【Abstract】 The objective of the current study was to optimize the fermentation conditions of producing fibrinolytic enzyme through single factor experiment and response surface experiment.The optimal fermentation conditions were obtained as follows:the carbon source was bran,the nitrogen source was yeast extract,the inorganic salt was 0.1% MgSO4,the solvent of medium was bean juice,pH was 4.0.Using 2% bran,2% yeast extract,0.1% MgSO4 as the major factor,choosing the right step,designing response surface experiment,the second-order regression model was obtained fitting the experimental data results.The R2 of the model was 0.8709,and was significant at the 90% of the probability.The second-order model predicted that under the conditions of 2.97% bran,2.30% yeast extract paste,0.147% MgSO4,pH 4,the highest fibrinolytic activity predicted was 410 IU/mL.
