北京市2007年从手足口病疫情标本中分离到人肠道病毒71型

【摘要】 目的对2007年5月底,在北京市大兴区发生的一起手足口病(Hand Foot Mouth Disease,HFMD)疫情进行实验室分析鉴定。方法采集HFMD患儿的疱疹液、咽拭子、粪便标本,采用逆转录-聚合酶链反应(Reverse Tran-scription-Polymerase Chain Reaction,RT-PCR)扩增标本及病毒阳性分离物中人肠道病毒(Human Enterovirus,HEV)特异性核酸片段,比较RT-PCR方法和HEV分离的敏感性。然后利用测序方法对HEV阳性分离物进行HEV分型鉴定,确定本次疫情中HEV的型别。结果5例HFMD患儿的10份标本中,有8份测到HEV特异核酸片段,检出阳性率为80%,高于HEV分离阳性率(30%)。5例患儿和9例密切接触者的共19份标本中共分离到5株HEV,其中来自2例患儿和1例密切接触者的4株病毒为EV71型(HEV71),来自密切接触者的另外1株HEV为柯萨奇病毒A组16型。结论北京市大兴区5月底发生的一起HFMD疫情是由HEV71型引起的。

【Abstract】 Objective Analyzing and identifying the type of enterovirus of human Hand Foot Mouth Disease(HFMD)outbreak in Daxing Bistrict in Beijing at the end of May in 2007. Methods The liquid of Herpes,throat swab and stool specimen were collected.Using reverse transcription-polymerase chain reaction(RT-PCR)method to amplify the enterovirus specific nucleotide acid fragment from specimens and virus positive cultures,the sensitivity of two methods was compared.Then identifying and typing of the enterovirus of HFMD through analyzing the results of nucleotide sequencing of the virus positive cultures. Results In 10 specimens from 5 HFMD children patients,enterovirus specific nucleotide acid fragment was detected in 8 patients,the RT-PCR positive ratio(80%) was higher than enterovirus isolation positive ratio(30%).In 5 enteroviruses isolated from 5 patients and 9 close contacts,4 isolated from 2 patients and 1 close contact were Enterovirus 71,1 isolated from close contact was Coxsackievirus A16. Conclusion Enterovirus 71 was the pathogen of HFMD outbreak in Daxing district at the end of May in 2007.
