辐射诱发胸腺淋巴瘤与Ikaros及p16基因多态性
Association between polymorphism of Ikaros and p16 genes and thymic lymphoma induced by ionizing radiation in mice
【摘要】 目的探讨Ikaros及p16基因多态性与辐射诱发小鼠胸腺淋巴瘤的相关性。方法采用Prom ega公司基因组DNA纯化试剂盒提取胸腺淋巴瘤细胞基因组DNA。采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)方法检测Ikaros基因rs28185870位点(C/T)、rs28185923位点(C/T)及p16基因rs3695947位点(A/G)单核苷酸多态性(SNPs)。结果C57BL/6 J及BALB/c小鼠rs28185870位点基因型分别为T/T、C/C;rs28185923位点基因型分别为C/C、T/T;rs3695947位点基因型分别为A/A、G/G。结论辐射敏感性不同的C57BL/6 J、BALB/c小鼠的rs28185870、rs28185923及rs3695947位点基因型不同,推测Ikaros及p16基因多态性与辐射诱发胸腺淋巴瘤相关。
【Abstract】 Objective To investigate the association between thymic lymphoma induced by ionizing radiation and the polymorphism of Ikaros and p16 genes in mice.Methods Genomic DNA of thymic lymphoma cells was extracted by genomic DNA purification kit.PCR-restriction fragment length polymorphism(RFLP) was used to examine the single nucleotide polymorphisms(SNPs) of rs28185870(C/T) and rs28185923(C/T) of Ikaros gene and rs3695947(A/G) of p16 gene.Results The genotype of rs28185870 site in C57BL/6J and BALB/c was T/T and C/C.The genotype of rs28185923 site was C/C and T/T.The genotype of rs3695947 site was A/A and G/G.Conclusion The genotypes of rs28185870,rs28185923 and rs3695947 in C57BL/6J and BALB/c mice with different radiosusceptibility were different.It is suggested that there is an association between the polymorphism of Ikaros and p16 genes and thymic lymphoma induced by ionizing radiation.
