蕨菜黄酮对小鼠急性乙醇肝毒性保护作用

【摘要】 目的研究蕨菜黄酮提取物对小鼠急性乙醇肝毒性的保护作用。方法60只小鼠随机分成乙醇模型组、空白对照组和低、中、高3个黄酮剂量组。3个剂量组连续灌胃45 d蕨菜黄酮后1次灌胃50%乙醇;乙醇模型组连续灌胃45 d纯净水后灌胃一次50%乙醇。空白对照组连续灌胃45 d纯净水。测定血清中谷丙转氨酶(GPT)活性、肝脏中甘油三酯(TG)和丙二醛(MDA)的浓度及肝脏指数。结果与乙醇模型组GPT(33.1546±13.5162),TG(0.1077±0.0291)相比,低剂量蕨菜黄酮提取物可降低血清GPT(26.9727±9.8373)、肝脏TG(0.0776±0.0281)水平(P<0.05);中剂量可降低血清GPT(17.3500±1.7656)(P<0.01)、肝脏TG(0.0669±0.0264)水平(P<0.01)和肝脏MDA(0.4493±0.1501)水平(P<0.05);高剂量可降低血清GPT(30.1714±16.4587)、肝脏TG(0.0751±0.0300)水平(P<0.05)和肝脏MDA(0.3897±0.0935)水平(P<0.01);空白对照组肝脏指数最低,蕨菜黄酮提取物未使增大的肝脏指数恢复到正常水平。结论蕨菜黄酮提取物对小鼠急性乙醇肝毒性有保护作用。

【Abstract】 Objective To explore the protective effect of Pteridium aquilinum flavanone extract on ethyl alcohol hepatic toxicity in mice and its possible mechanism.MethodsSixty mice were random divided into 5 groups: low,middle and high dose groups,alcohol model group and normal control group.After gavaged with flavanone extract for 45 days,the low,middle and high dose groups were gavaged with 50% ethyl alcohol.The ethyl alcohol model group were first gavaged with pure water continuously for 45 days,then gavaged with 50% ethyl alcohol.The normal control group were gavaged with pure water for 45 days.The GPT in blood serum,the TG and the MDA in liver and the liver index were determined by colorimetric analysis.ResultsCompared with the ethyl alcohol model group,low dose Pteridium aquilinum flavanone could obviously reduce the GTP in blood serum and the TG in liver(P<0.05).Middle dose Pteridium aquilinum flavanone extract obviously reduced the GTP in blood serum and the TG in liver(P<0.01)and the MDA in liver(P<0.05).High dose Pteridium aquilinum flavanone extract obviously reduced the GPT in blood serum and the TG in liver(P<0.05)and reduced the MDA in liver(P<0.01).The liver index of the normal control was the lowest.The Pteridium aquilinum flavanone extract could not change the increased liver index to normal level.ConclusionPteridium aquilinum flavanoe extract has protective effect on ethyl alcohol hepatic toxicity in mice.
