3-硝基丙酸预处理对大鼠缺血-再灌注脑损伤保护作用的研究
Protective effect of 3-nitropropi onic acid preconditioning on focal cerebral ischemia/reperfusion in rats
【摘要】 目的研究3-硝基丙酸(3-NPA)预处理对大鼠局灶性脑缺血脑梗死体积,脑缺血半暗带神经细胞凋亡的影响,探讨3-NPA预处理诱导脑缺血耐受的机制。方法大鼠腹腔注射3-NPA,分别在24h和72h后制作大脑中动脉缺血-再灌注模型,采用TTC染色和TUNEL法,观察3-NPA预处理对缺血2h再灌注24h脑梗死体积、神经细胞凋亡的影响。结果缺血2h再灌注24h,3-NPA预处理组脑梗死体变小,神经细胞凋亡减少,与对照组及假手术组比较有显著性差异。结论3-NPA预处理可以诱导脑缺血耐受,抑制神经细胞凋亡可能是其机制之一。
【Abstract】 Objective To discuss the effects of 3-nitropropi onic acid(3-NPA)preconditioning on the volume of infarction of focal cerebral ischemia in rats and on the neuronal apoptosis of cerebral ischemic penumbra,and to clarify the mechanism of ischemic tolerance induced by 3-NPA preconditioning.Methods Animal models of middle cerebral artery ischemia-reperfusion were performed 24h and 72h after the intraperitoneal injection of 3-NPA.TTC staining and TUNEL assay were adopted.The effects of 3-nitropropionic acid preconditioning on the volume of cerebral infarction and neuronal apoptosis caused by ischemia for 2 hours and reperfusion for 24 hours was evaluated.Results The 3-NPA pretreatment group had smaller infarct volume and fewer neuronal apoptosis after 2h ischemia and 24h reperfusion,and showed significant difference with the control group and sham operation group.Conclusion 3-NPA preconditio ning can induce cerebral ischemic tolerance,and the inhibition of neuronal apoptosis may be one of the mechanisms.
