鼻息肉中STAT3和VEGF的表达与相关性研究
Expression and correlation of STAT3 with VEGF in the nasal polyps
【摘要】 目的:检测信号转导及转录活化因子3(signal transducers and activators of transcription3,STAT3)、磷酸化信号转导及转录活化因子3(phosphor-STAT3,p-STAT3)及血管内皮生长因子(vascular endothelial growth factor,VEGF)在鼻息肉组织中的表达及分布,探讨他们在鼻息肉发病机制中的作用及其相关性。方法:选取符合纳入标准的鼻息肉患者手术切除标本30例,选取同期单纯行鼻中隔偏曲矫正术中切除的下鼻甲组织10例。采用免疫组化链霉菌抗生物素蛋白-过氧化物酶(SP)法检测30例鼻息肉标本(鼻息肉组)与下鼻甲黏膜(对照组)中STAT3、p-STAT3及VEGF的表达。结果:STAT3在鼻息肉中粘膜上皮及腺体的表达的阳性率为66.67%,鼻息肉组STAT3表达明显高于下鼻甲组33.33%(P<0.01)。p-STAT3在鼻息肉中粘膜上皮及腺体的表达的阳性率为56.67%,鼻息肉组p-STAT3表达明显高于下鼻甲组43.33%(P<0.01)。VEGF在鼻息肉中粘膜上皮及腺体的表达的阳性率为76.67%,鼻息肉组VEGF表达明显高于下鼻甲组23.33%(P<0.01)。p-STAT3的表达与VEGF的表达之间呈正相关(r=0.48,P<0.01)。结论:鼻息肉组织中p-STAT3与VEGF表达呈正相关,提示VEGF基因可能由p-STAT3蛋白调节,STAT3、p-STAT3及VEGF的表达可能是鼻息肉发病机制中起重要作用的因素之一。
【Abstract】 AIM:To detect the expression and distribution of signal transducers and activators of transcription3,phosphor-STAT3 and vascular endothelial growth factor in nasal polyps,and to investigate the role of STAT3,phosphor-STAT3 with VEGF in the pathogenetic mechanism of nasal polyps.METHODS:All selected cases were measured up the standard of enrollment.30 samples of nasal polyps were obtained from patients undergoing nasal polypectomy,and 10 samples of inferior turbinate tissues were from the patients undergoing nasal septal construction.STAT3,phosphor-STAT3 and VEGF in nasal polyp tissues from 30 nasal polyposis patients and 10 samples of inferior turbinate tissues were detected by immunohistochemistry method.RESULTS:In nasal polyp tissues,the positive rates of STAT3,p-STAT3 and VEGF expressions were 66.67%,56.67% and 76.67% respectively,all were significantly higher than those in control inferior turbinates(P<0.01).A positive correlation between the expression of p-STAT3 and that of VEGF(r=0.48,P<0.01) was observed.CONCLUSION:There is significant positive correlation between the expression of VEGF and p-STAT3 in the tissues of nasal polyps,suggesting that VEGF gene may be regulated directly by p-STAT3 protein.The expression of STAT3,p-STAT3 and VEGF in nasal polyps may play an important role in the pathogenetic mechanism of nasal polyps.
