血小板反应蛋白1在转化生长因子β1诱导的大鼠心肌成纤维细胞中的作用
Effects of thrombospondin 1 on rat cardiac fibroblasts induced by transforming growth factor β1
【摘要】 目的:观察血小板反应蛋白1(TSP-1)在转化生长因子β1(TGF-β1)诱导大鼠心肌成纤维细胞中的作用。方法:差速贴壁法分离新生Sprague-Dawley(SD)大鼠心肌成纤维细胞(CFs)。MTT法测CFs生长,羟脯氨酸法测CFs胶原含量,RT-PCR法测TSP-1 mRNA表达。结果:TGF-β1诱导CFs TSP-1的表达呈浓度-时间依赖性增加(P<0.01)。TGF-β1(20μg/L)作用下24 h可诱导CFs增殖和胶原合成(P<0.01)。TSP-1反义寡核苷酸作用后的CFs的MTTA值、胶原含量减少(P<0.01)。结论:TGF-β1可诱导新生大鼠CFs TSP-1表达,TSP-1反义寡核苷酸对TGF-β1诱导大鼠CFs增殖与胶原合成具有明显的抑制作用,TSP-1在TGF-β1诱导大鼠心肌纤维化中可能具有重要的作用。
【Abstract】 AIM: To investigate the effects of thrombospondin 1 on transforming growth factor β1 induced rat cardiac fibroblasts(CFs).METHODS: CFs of neonatal Sprague-Dawley(SD) rats were isolated with the method of digestion and differential anchoring velocity.The proliferation and collagen synthesis of rat CFs were observed with MTT and hydroxyproline.The expression of TSP-1mRNA was analyzed by RT-PCR.RESULTS: The dose and time-dependent effects of TGF-β1 were observed.Expression of TSP-1 was increased significantly(P<0.01).Stimulation of CFs with TGF-β1(20 μg/L,24 h) significantly increased CFs proliferation and collagen synthesis(P<0.01).TSP-1 antisense oligonucleotide effectively inhibited TGF-β1 induced CFs proliferation and collagen synthesis(P<0.01).CONCLUSION: The proliferation and collagen synthesis of CFs induced by TGF-β1 are inhibited by TSP-1 antisense oligonucleotide,which may exert helpful effect on anti-fibrosis.
