茶多酚对肠缺血-再灌注后大鼠肺损伤的影响

【摘要】 目的观察茶多酚对肠缺血-再灌注所致肺损伤的作用,考察茶多酚对髓过氧化物酶(MPO)和肿瘤坏死因子-α(TNF-α)的影响,探讨茶多酚防治肠源性肺损伤的机制。方法将32只雄性SD大鼠随机分为4组各8只:I/R组:暴露腹腔后夹闭肠系膜上动脉(SMA)1h,开放再灌注2h;茶多酚预处理组(P1组):肠缺血前10min给予茶多酚;茶多酚治疗组(P2组):肠再灌注前10min给予茶多酚;假手术组:仅暴露SMA,不行肠I/R及茶多酚输注。茶多酚剂量为首剂10mg·kg-1,然后以10mg·kg-1.h-1持续输注。所有大鼠于再灌注2h处死,检测血浆与肺组织MPO、TNF-α含量,制作肺与空肠病理切片。结果肠I/R后大鼠血浆、肺组织MPO、TNF-α表达明显增加。而茶多酚可以抑制上述改变,以P1组效果最为明显。结论MPO、TNF-α在肠I/R后肺损伤的发生中发挥重要作用。肠I/R早期应用茶多酚可明显减少血浆和肺组织MPO、TNF-α表达,在一定程度上减轻肺损伤。

【Abstract】 Objective To explore the protective effect of tea polyphenols on lung injury after intestinal ischemia/reperfusion(I/R)in rats,and the potential mechanism of which.Methods Thirty two male SD rats were randomly divided into 4 groups(n=8 in each).①Group I/R,in which rats were subjected to 1 hour of occlusion of the superior mesenteric artery(SMA),followed by 2 hours of reperfusion;② Early treatment group(P1),rats were subjected to the same procedure as group I/R with the additional administration of tea polyphenols 10 minutes before ischemia at 10 mg·kg-1 loading dose,followed by continuous infusion at 10 mg·kg-1·h-1;③ Treatment group(P2),rats were subjected identical treatment as group I/R with the administration of tea polyphenols started 10 minutes before reperfusion at 10 mg·kg-1 loading dose,followed by continuous infusion at 10 mg·kg-1·h-1;④Sham-operation group,rats were subjected to laparotomy only,but received normal saline at 10 mL·kg-1·h-1.At the end of reperfusion,all animals were sacrificed.The MPO and TNF-α in plasma and lung tissue was assessed.Pathological sections of lung and empty intestine were made Results MPO and TNF-α in plasma and expression in lung tissue were increased significantly in group I/R.All the increase was less in quantity in the early treatment group of tea polyphenols than those in the other two groups,and there was significant difference in contents and expression in lung between group I/R and group P1(P<0.05).Conclusion MPO and TNF-α plays an important role in lung injury after intestinal I/R.The early treatment of tea polyphenols before intestinal I/R may be more beneficial by reducing MPO and TNF-α expression in lung injury.
