环孢菌素A合成酶及其遗传改造研究进展
Advancement in researches of cyclosporine A and its genetic modification
【摘要】 环孢菌素A是从真菌中提取的一种新型免疫抑制剂,可选择性作用于T细胞,抑制其活化,现主要应用于器官移植时的排斥反应。环孢菌素A是由环孢菌素A合成酶通过模块指导依次合成并环化成环肽。环孢菌素家族已有2000多种,应用到免疫,抗病毒肝炎等许多领域。本文简要介绍环孢菌素A合成酶的结构及合成环孢菌素A的机制,以及对其实施遗传工程改造方面的研究进展。
【Abstract】 Cyclosporine A is a new immunosuppressant extracted from Tolypocladium inflatum which selectively inhibits T cells activation and is primarily applied in rejection of organ transplantation.The biosynthetic modules of cyclosporine synthetase assemble cyclosporine A to cyclic peptides.The cyclosporine family contains over 2000 members and is involved in immunomodulatory reaction, anti-HCV effects and so on.Here we discuss the recent advances in CssA,including the advances in the structure of cyclosporine synthetase,the synthesis mechanisms of cyclosporine and the modification of the CssA gene.
