雌二醇水平降低加速大鼠实验性牙周炎的发展
Effects of Estradiol on the Chronic Periodontitis in Experimental Rat
【摘要】 目的探索雌二醇水平与大鼠牙周炎发展间的关系。方法24只雌性Wister大鼠随机分为3组:对照组,假手术组和去势组。利用钢丝结扎牙齿的方法对去势组和假手术组大鼠构建慢性牙周炎动物模型,通过检测血清雌二醇水平,观察X线片和HE切片,同时比较各组大鼠牙周组织中MMP-9的表达量来综合评价各组牙周炎的严重情况。结果与对照组和假手术组比较,去势组雌二醇水平明显降低(P<0.01)。X线片观察发现假手术组结扎侧第二磨牙近远中和根分歧处有明显的牙槽骨吸收,而去势组的骨吸收较假手术组更为严重。组织形态学观察发现去势组结合上皮中有大量中性粒细胞浸润,牙槽骨吸收明显,而假手术组的牙周炎症状不如去势组严重。免疫组化结果显示去势组第二磨牙牙龈及牙周膜中MMP-9呈强阳性表达,而假手术组MMP-9的表达要弱于去势组,对照组的阳性细胞的表达最少。结论雌激素水平降低促进了牙周组织的破坏,而且MMP-9参与了牙周组织的破坏过程。
【Abstract】 Objective In this research,the relationship between blood estrogen level and periodontitis in rats was studied. Methods Twenty-four female Wister rats were randomly divided into three groups:control group,sham operation group and ovariectomized group. Rat chronic periodontitis model was established by ligating the rat’s second molar with a steel wire in the sham operation group and ovariectomized group. Serum estrogen level of the rats was detected respectively. Dental X-ray observation,histological examination and immunohistochemistry were employed for the characterization of rat’s chronic periodontitis. Results Compared with both control group and sham operation group,a significant decrease in scrum estrogen level was observed in ovariectomized group (P<0.01). X-ray observation indicated that severe alveolar bone absorption happened to the ovariectomized group. Massive neutrophils infiltration in unctional epithelium was also confirmed by histological examination. However,the above mentioned chronic periodontitis symptom was not so significant in the rats of sham operation group. While MMP-9,as examined by immunohistochemistry,was revealed as strong positive in the second molar’s gingival tissues and periodontitis ligament of the rats in ovariectomized group. Expression of MMP-9 was much lower in sham operation group and even fewer in control group. Conclusion Lower estrogen level plays an improving role in the development of periodontitis.MMP-9 may participate in the periodontal tissue destruction.
