IGF-1与survivin在反应性和肿瘤性星形胶质细胞中的表达及意义

【摘要】 目的探讨IGF-1、Bcl-2、survivin和Ki-67蛋白在星形胶质细胞反应性增生与低级别星形细胞瘤中表达的区别及意义。方法应用组织芯片和免疫组化染色技术检测正常脑组织、星形胶质细胞反应性增生、低级别(Ⅰ~Ⅱ级)和高级别(Ⅲ~Ⅳ级)星形细胞瘤中IGF-1、Bcl-2、survivin和Ki-67蛋白的表达。结果正常脑组织中4种指标均为阴性表达;IGF-1、Bcl-2、survivin和Ki-67在星形胶质细胞反应性增生组中阳性率分别为28.9%、26.7%、26.7%、22.2%;在低级别星形细胞瘤组中阳性率分别为63.8%、50.0%、53.2%、70.2%;在高级别星形细胞瘤组中阳性率分别为88.9%、79.2%、88.1%、95.2%。IGF-1、Bcl-2、survivin和Ki-67在各实验组间比较均差异显著(P<0.01);他们的表达与组织学分级密切相关(P<0.01);他们相互之间的相关性检验呈密切相关(P<0.01)。结论IGF-1、Bcl-2、survivin和Ki-67在星形胶质细胞反应性增生与低级别星形细胞瘤的鉴别诊断中可能会有一定的意义;IGF-1、Bcl-2和survivin在星形细胞瘤的发生、发展中可能起重要作用。

【Abstract】 Objective To study the difference of IGF-1,Bcl-2,survivin and Ki-67 protein expression in reactive and neoplastic astrocytes and the significance of them.Methods Immunohistochemistry and tissue microarray techniques were used to determinate the expression of IGF-1,Bcl-2,survivin and Ki-67 protein in normal brain tissues, astrocytes proliferation,low-grade astrocytomas, and high-grade astrocytomas.Results The expression of IGF-1,Bcl-2,survivin and Ki-67 protein were all negative in control group.The positive expression rates of them in reactive astrocytes were 28.9%, 26.7%, 26.7% and 22.2%,respectvely; and in low-grade astrocytomas were 63.8%, 50.0%, 53.2%,70.2%; in high-grade astrocytomas were 88.9%, 79.2%, 88.1%,95.2%.IGF-1,Bcl-2,survivin and Ki-67 all had significant difference among the three experimental groups(P<0.01); the expression of them had close correlation to the histological grades ( P<0.01),had close correlation each other(P<0.01).Conclusion IGF-1,Bcl-2,survivin and Ki-67 protein might be useful for discriminating between reactive astrocytes proliferation and low-grade astrocytomas, IGF-1、survivin and Bcl-2 protein might play an important role in the progression of astrocytomas.
