包裹光敏剂m-THPC的PLGA微球的体外释放特性研究

【摘要】 目的:测量包裹光敏剂m-THPC的PLGA微球的载药量,并定量分析在含不同浓度脂肪乳和血清的水溶液中PLGA微球的体外释放规律。方法:称取一定质量PLGA微球,置入一定体积的二氯甲烷和甲醇混合有机溶剂中完全溶解,测量溶液荧光强度,与荧光强度—光敏剂m-THPC浓度标准曲线对比得出光敏剂m-THPC的浓度后计算出PLGA微球的载药量;称取一定质量的载药量已知的PLGA微球,置入200mL含脂肪乳和血清的水溶液中释放,37℃恒温摇床摇动,每次取样0.5mL,测量溶液荧光强度,与荧光强度—光敏剂m-THPC浓度标准曲线对比得出光敏剂m-THPC的浓度后计算出光敏剂m-THPC的释放量。结果:血清能加速PLGA微球的释放,同时脂肪乳浓度越高,PLGA微球释放光敏剂m-THPC的速度越快。结论:给病人注射适量的脂肪乳注射液,有可能提高PLGA微球释放速度和光敏剂m-THPC在血液中的浓度,从而提高光动力治疗效率。
【Abstract】 Objective: Determination of the drug loading of the PLGA microparticles enwrapping photosensitizer m-THPC,and quantitative analysis of the m-THPC release in vitro of these PLGA microparticles in water solution containing intralipid and serum. Methods: Dissolving some PLGA microparticles in the mixture solution of Methylene Dichloride and Methyl Alcohol, and the concentration of m-THPC in the solution is determined by comparing the fluorescence intensity of the solution with the prefabricated standard intensity-concentration curve,thus the drug loading of the PLGA microparticles can be calculated. Dis- solving PLGA microparticles whose drug loading is known in 200 mL water solution of intralipid and serum, and stir the solution at 37℃. Taking 0.5 mL sample each time from the solution, and the concentration of m-THPC is determined by comparing the fluorescence intensity of the sample with the prefabricated standard intensity-concentration curve, so the quantity of the m-THPC released from these PLGA microparticles can be calculated. Results: The m-THPC release rate of these PLGA microparticles can be accelerated by increasing the concentration of intralipid or serum. Conclusion: The m-THPC release rate of these PLGA microparticles in the blood of the patient can be accelerated if the patient has been injected with some intralipid. This will im- prove the concentration of m-THPC in the blood as well as the efficiency of the photo dynamic therapy on the patient.
